不同艾灸量对寒凝证类痛经大鼠疼痛反应及脊髓内μ受体mRNA表达的影响

【摘要】:目的:观察不同刺激量艾灸关元穴对寒凝证类痛经大鼠扭体反应及脊髓内μ受体表达的影响,探讨不同灸量的镇痛效应及其部分中枢机制。方法:按随机数字表法将60只雌性SD大鼠,随机分为盐水组、寒凝证类痛经模型组、A组(艾灸1壮)、B组(艾灸3壮),每组15只。除盐水组外,其它各组采用全身冷冻法结合苯甲酸雌二醇注射液加缩宫素注射法制备寒凝证类痛经大鼠模型,A组和B组以不同灸量艾灸关元穴。观察大鼠扭体反应,并以荧光定量PCR(RT-PCR)法测定脊髓内μ受体mRNA的表达水平。结果:(1)扭体反应:与盐水组比较,模型组、B组扭体潜伏期缩短、扭体次数、扭体评分明显增高(P0.05)。与模型组比较,A组、B组扭体次数、扭体评分明显减少(P0.05)。(2)脊髓μ受体mRNA的表达:与盐水组比较,A组与B组的表达均增强(P0.01),与模型组相比,A组与B组的表达增强(P0.01)结论:(1)不同艾灸量施予关元穴对寒凝证类痛经大鼠模型均有镇痛作用。(2)艾灸产生镇痛效应的部分机制可能通过增强寒凝证类痛经模型大鼠脊髓内阿片受体的表达产生镇痛效应。
[Abstract]:Aim: to observe the effects of moxibustion at Guanyuan point on writhing reaction and expression of 渭 receptor in spinal cord of rats with dysmenorrhea with cold coagulation syndrome, and to explore the analgesic effect of different amounts of moxibustion and some central mechanisms. Methods: according to the method of random digital table, 60 female SD rats were randomly divided into saline group, cold coagulation syndrome dysmenorrhea model group and group A (moxibustion 1 strong), B group (moxibustion 3 strong) with 15 rats in each group. Except saline group, the rat model of cold coagulation syndrome dysmenorrhea was established by whole body freezing combined with estridiol benzoate injection and oxytocin injection. Group A and group B were treated with different amounts of moxibustion at Guanyuan point. The writhing reaction of rats was observed, and the expression of 渭 receptor mRNA in spinal cord was measured by fluorescence quantitative PCR (RT-PCR). Results: (1) writhing reaction: compared with saline group, the latency of writhing was shortened, the number of writhing and the score of writhing in group B were significantly higher than those in saline group (P 0.05). Compared with the model group, the number of writhing and the score of writhing in group A and group B were significantly lower than those in group B (P 0.05). (2). The expression of mRNA in spinal cord 渭 receptor in group A and group B was significantly higher than that in group B (P 0.01), and that in group A and group B was significantly higher than that in group B (P 0.01). The expression of group A and group B was increased (P 0.01) conclusion: (1) different doses of moxibustion at Guanyuan acupoint have analgesic effects on the rat model of dysmenorrhea with cold coagulation syndrome. (2) part of the mechanism of analgesic effect of moxibustion may have analgesic effect by increasing the expression of opioid receptor in spinal cord of rats with cold coagulation syndrome dysmenorrhea.
