脾虚痰浊证巴马小型猪心肌糖代谢相关分子的变化特点

【摘要】:目的:探讨脾虚痰浊证巴马小型猪心肌组织葡萄糖代谢相关分子的变化特点。方法:普通级雄性巴马小型猪10只随机分为正常组及脾虚痰浊组,每组5头。采用疲劳过度联合高脂单笼饲养建立小型猪脾虚痰浊证模型。采用实时荧光定量PCR法检测心肌组织葡萄糖转运体4(glucose transporter 4,GLUT4)、磷酸果糖激酶1(Phosphofructokinase-1,PFK-1)、丙酮酸激酶(Pyruvate kinase2,PKM2)和己糖激酶(Hexokinase,HK)mRNA表达,应用Western blotting法检测心肌组织GLUT4、PFK1、PKM2和HK蛋白表达水平。结果:与正常组小型猪相比,脾虚痰浊组小型猪心肌组织GLUT4、PKM2、PFK1、HK mRNA表达水平显著下降(P0.01),GLUT4、PKM2蛋白表达水平显著下降(P0.05)。PFK1、HK蛋白表达水显著下降P0.01)。结论:脾虚痰浊证小型猪心肌存在葡萄糖代谢异常,与葡萄糖转运及代谢相关分子表达呈下降趋势。
[Abstract]:Objective: to investigate the changes of glucose metabolism molecules in myocardium of Bama miniature pigs with spleen deficiency and phlegm turbid syndrome. Methods: ten male Bama miniature pigs of common grade were randomly divided into normal group (n = 5) and spleen deficiency and phlegm turbid group (n = 5). The model of spleen deficiency and phlegm turbid syndrome of miniature pigs was established by fatigue combined with hyperlipidemia and single cage feeding. The expression of glucose transporter 4 (glucose transporter 4, glut 4, phosphofructokinase 1 (Phosphofructokinase-1,PFK-1), pyruvate kinase (Pyruvate kinase2,PKM2) and hexokinase (Hexokinase,HK) mRNA in myocardial tissue was detected by real time fluorescence quantitative PCR, and the expression of GLUT4,PFK1,PKM2 and HK protein in myocardial tissue was detected by Western blotting method. Results: compared with the normal group, the expression level of GLUT4,PKM2,PFK1,HK mRNA and GLUT4,PKM2 protein in myocardial tissue of miniature pigs with spleen deficiency and phlegm turbid group were significantly lower than those in normal group (P 0.05). The expression of PFK1 and HK protein was significantly decreased by P 0.01). Conclusion: there is abnormal glucose metabolism in the myocardium of miniature pigs with spleen deficiency and phlegm turbid syndrome, and the expression of glucose transport and metabolism related molecules is decreased.
