蒙药森登-4复方及其单味药的多糖成分提取及抗氧化作用研究

【摘要】:多糖是蒙药森登-4及其各单味药的重要活性成分之一。具有增强人体免疫、降血脂、降血糖、抗肿瘤、抗细菌、抗病毒、抗辐射、抗凝血、抗衰老、抗氧化等作用。本论文首先对森登-4及其各单味药中多糖的提取工艺进行研究,试验中以多糖含量为指标,以固液比、提取时间、提取温度和提取次数为因素,使用响应面设计优化了蒙药森登-4复方及其单味药的多糖最佳水提工艺;其次,探讨了蒙药森登-4复方及其各单味药中多糖对羟基自由基(·OH-)、超氧阴离子自由基(O2-)、亚硝酸根离子(NO2-)和DPPH自由基的清除能力以及多糖的总还原能力,从中探讨蒙药森登-4配伍前后抗氧化活性的变化。多糖提取工艺的研究结果表明,森登-4及其单味药的最佳提取条件各不相同。(1)森登-4多糖的最佳提取工艺为:固液比1:40,提取温度为95℃,提取时间为3 h,提取次数为3次,多糖提取百分含量为8.90%。(2)文冠木多糖最佳提取工艺为:固液比1:32,提取温度为69℃,提取时间为1.8 h,提取次数为2次,多糖提取百分含量为1.45%。(3)川楝子多糖的最佳提取工艺为:固液比1:20,提取温度为90℃,提取时间为2.9 h,提取次数为3次,多糖提取百分含量为22.34%。(4)栀子多糖的最佳提取工艺为:固液比1:33.7,提取温度为80℃,提取时间为3 h,提取次数为3次,多糖提取百分含量为10.97%。(5)诃子多糖的最佳提取工艺为:固液比1:28,提取温度为69℃,提取时间为1 h,提取次数为3次,多糖提取百分含量为5.99%。多糖抗氧化研究表明,配伍前后药材多糖的抗氧化活性发生变化,且森登-4多糖的抗氧化活性优于其四种单味药。(1)森登-4多糖清除·OH-的能力强于对照品Vc、文冠木、栀子、川楝子,弱于诃子。其中栀子清除羟基自由基的能力达26.35%。(2)森登-4多糖清除O2-的能力强于对照品甘露醇和其他四种药材。(3)森登-4多糖清除NO2-的能力强于Vc、诃子、川楝子,弱于栀子,与文冠木相当。(4)川楝子和诃子清除DPPH的能力弱于Vc。森登-4、文冠木和栀子多糖对DPPH的清除作用未能测试到。(5)蒙药森登-4及其各单味药中多糖的还原能力弱于对照品Vc的还原能力,且森登-4的还原能力也弱于其他四中单味药。
[Abstract]:Polysaccharide is one of the important active components of Mongolian medicine Sendeng-4 and its single drugs. It has the functions of enhancing human immunity, lowering blood lipid, lowering blood glucose, anti-tumor, anti-bacterial, anti-virus, anti-radiation, anticoagulant, anti-aging, antioxidant and so on. In this paper, the extraction technology of polysaccharide from Sendeng-4 and its single drug was studied. In the experiment, the optimal water extraction process of polysaccharide from Sendeng-4 compound and its single drug was optimized by using response surface design, taking the content of polysaccharide as the index, the ratio of solid to liquid, extraction time, extraction temperature and extraction times as factors. Secondly, the scavenging ability of polysaccharides to hydroxyl radical (OH -), superoxide anion radical (O2 -), nitrous acid ion (NO2-) and DPPH radical and the total reduction ability of polysaccharide in Mongolian medicine Sendeng-4 compound and its single drugs were discussed, and the changes of antioxidant activity before and after the compatibility of Mongolian medicine Sendeng-4 were discussed.
