牛鞭草属DUS测试材料的SSR指纹图谱构建

【摘要】:高效快速的品种(系)DNA指纹图谱鉴定,可为辅助牛鞭草属品种(系)DUS测试和知识产权保护等提供科学依据。本研究从54对SSR引物中筛选出20对引物对牛鞭草属25个DUS测试材料进行鉴定分析。结果表明,20对SSR引物对牛鞭草属25个DUS测试材料共扩增出253条清晰条带,其中多态性条带241条,多态性条带比率(PPB)为95.26%。每对引物的多态信息含量(PIC)为0.599~0.900,平均为0.760。UPGMA聚类分析表明,牛鞭草属25个材料在相似系数为0.668处可分为三大类群。20对SSR引物能鉴别10~22个牛鞭草属材料,引物DHSSR7、引物DHSSR11和引物DHSSR15的有效组合可鉴别所有供试材料。以3对SSR引物扩增的电泳图谱为基础,建立了牛鞭草属25个DUS测试材料的指纹图谱标准模式图,能够快速区分各个供试材料。
[Abstract]:The high efficiency and rapid identification of DNA fingerprinting can provide scientific basis for assisting DUS testing and intellectual property protection of bullwhip varieties (lines). In this study, 20 pairs of SSR primers were screened out from 54 pairs of primers to identify and analyze 25 DUS test materials of the genus Verbena. The results showed that a total of 253clear bands were amplified by 20 pairs of SSR primers against 25 DUS materials, of which 241were polymorphic bands, and the ratio of polymorphic bands was 95.26%. The polymorphic information content (PIC) of each pair of primers was 0.599 ~ 0.900, and the average 0.760.UPGMA cluster analysis showed that: Twenty-two pairs of SSR primers could be used to identify 10 and 22 materials, and the effective combination of primer DHSSR7, primer DHSSR11 and primer DHSSR15 could be used to identify all of the tested materials. The similarity coefficient of the 25 materials could be divided into three groups at the similarity coefficient of 0.668. 20 pairs of primers could be used to identify 10 to 22 materials of the genus Botrytis. Based on the electrophoresis patterns amplified by three pairs of SSR primers, the standard patterns of fingerprint patterns of 25 DUS materials of the genus bullwhip were established, which could quickly distinguish the tested materials from each tested material.
