罗非鱼胶原肽中试车间设计与中试生产

【摘要】:胶原肽不仅分子量小易于人体消化吸收,而且具有独特的生物活性,主要表现为抗高血压、抗氧化、抗肿瘤等方面,因此在医疗、保健及美容领域具有良好的应用前景。本研究立足于企业的需求,设计一个符合保健品生产要求的胶原肽GMP中试车间,并以罗非鱼胶原蛋白胨为原料,结合生物酶解技术、膜分离技术等现代生物技术在该GMP中试车间中制备具有生物活性的胶原肽。首先,设计一个胶原肽GMP中试车间。根据罗非鱼胶原肽的特点进行中试工艺论证,并在此基础上完成物料衡算、设备选型和水电用量估算等。该项目产量盈亏平衡点为49.3吨,经营安全率达52.1%,投资回收期为4.4年。在完成设计的基础上,绘制车间平面分布图、人流物流分布图、设备流程图和设备平面布置图。其次,以罗非鱼胶原蛋白胨为研究对象,采用不同酶解工艺,研究发现利用对虾加工副产物制备所得虾头酶的最佳酶解条件为:加酶量0.5%,酶解温度37℃,反应p H为8.0,酶解时间为3 h。碱性蛋白酶的酶解条件为:加酶量为0.1%,酶解温度45℃,反应p H是9.0,酶解时间为3 h。同时对两种酶解液进行分子量测定,发现酶解液的分子量相近,主要分布在2 k Da以下。根据实验优化后的酶解条件,进行罗非鱼胶原肽中试生产工艺探究,最终确立的工艺流程是:酶解、微滤、超滤、纳滤、喷雾干燥、包装。同时,中试生产过程必须遵循GMP相关标准,严格控制各个操作步骤的参数及卫生条件要求。而中试生产膜分离过程中随着时间的延长,膜污染增大,膜通量逐渐下降。最后,本文对罗非鱼胶原肽中试产品进行性质分析。对中试膜分离产生的各组分进行生物活性分析,虾头酶酶解制备的罗非鱼胶原肽具有较高的ACE抑制活性(IC50=0.52mg/m L),而碱性蛋白酶制备的罗非鱼胶原肽具有较高的抗氧化活性,其DPPH自由基清除率的IC50=5.7 mg/m L。这两种蛋白酶制备的胶原肽各组分分子量相近,均分布在1 k Da以下。另外,文中对虾头酶制备的胶原肽中试产品进行性质分析。产品感官特点是白色、无味、无肉眼可见杂质。在胶原肽的功能特性方面,其吸水性、持水性和溶解性较高,吸油性低。罗非鱼胶原肽产品模拟胃肠液消化后ACE抑制活性稍有降低(IC50=0.85mg/m L),但其体外抗高血压活性仍然较高。本文利用胶原肽GMP中试车间实现罗非鱼胶原肽的中试生产,对中试产品进行相关性质分析。这些研究为高生物活性罗非鱼皮胶原肽的产业化生产奠定坚实的基础,为罗非鱼加工副产物的高效利用提供有益参考,也为相关功能性食品的开发提供科学依据。
[Abstract]:Collagen peptide is not only easy to digest and absorb, but also has unique biological activity, such as anti-hypertension, anti-oxidation, anti-tumor and so on, so it has a good application prospect in medical, health care and beauty fields. Based on the demand of enterprises, a pilot workshop of collagen peptide GMP was designed to meet the requirements of health products production. The collagen peptone of tilapia was used as raw material, and the bioenzymatic hydrolysis technology was used. Membrane separation and other modern biotechnologies were used to prepare bioactive collagen peptides in the GMP pilot plant. First, a collagen peptide GMP pilot workshop was designed. According to the characteristics of collagen peptide of tilapia, the pilot-scale technological demonstration was carried out, and the material balance, equipment selection and the estimation of water and electricity consumption were completed on this basis. The profit and loss breakeven point of the project is 49.3 tons, the operating safety rate is 52.1 and the investment recovery period is 4.4 years. On the basis of completion of the design, draw workshop plane distribution map, flow distribution map, equipment flow chart and equipment layout plan.
