术中甲状旁腺体外临时保存的细胞活性影响因素研究

【摘要】:目的 研究甲状旁腺即时自体移植过程中,甲状旁腺组织术中体外临时保存的细胞活性影响因素;以期找到一种最佳的术中甲状旁腺体外临时保存条件,使甲状旁腺组织的活性影响降至最低,以进一步提高甲状旁腺自体移植的成功率。 方法 2014年01月至2014年12月,甲状腺癌手术中即刻切取12枚因受肿瘤侵犯而需要一并切除的新鲜甲状旁腺组织,每个切成7份小组织块;随机单盲分组,1份组织块直接新鲜固定,作为对照组,其余6份分别在室温生理盐水(22-24℃)和4℃DMEM培养液中保存0.5小时、1.0小时和2.0小时;最后所有组织块固定切片,行透射电子显微镜检查,观察细胞超微结构变化,评估细胞活性情况。 结果 ①新鲜甲状旁腺组织,在室温生理盐水中保存0.5小时后可见染色质固缩,少量线粒体肿胀变圆;保存1.0小时后可见核溶解、染色质凝集、核内空泡化,线粒体肿胀变大变圆、空泡样变性。 ②新鲜甲状旁腺组织,在4℃DMEM培养液中保存0.5小时后甲状旁腺细胞超微结构未见明显变化;保存1.0小时后,可见细胞核变大变圆,线粒体肿胀,空泡样变性。 结论 甲状旁腺体外临时保存的介质和保存时间长短是甲状旁腺细胞活性的影响因素,在甲状旁腺即时自体移植过程中最好采用4℃DMEM等细胞培养液作为体外临时保存溶液,保存时间不应超过1.0小时,越快行自体移植越好。
[Abstract]:Objective to study the factors affecting the cell viability of parathyroid tissue during the immediate autotransplantation of parathyroid gland in vitro, and to find out the best condition for the preservation of parathyroid gland in vitro. The activity of parathyroid tissue was minimized to further improve the success rate of autotransplantation of parathyroid gland. Methods from January 2014 to December 2014, 12 fresh parathyroid tissues, which needed to be removed simultaneously because of tumor invasion, were removed immediately during the operation of thyroid carcinoma, each of which was cut into 7 small tissue blocks.
