有氧运动对MI后大鼠心肌细胞端粒酶活性及细胞增殖的影响研究

【摘要】:研究目的:探讨有氧运动对心肌梗死(MI)后大鼠心肌细胞端粒酶活性及心肌细胞增殖的影响。研究方法:选用雄性SD大鼠30只,随机分为假手术组(sham),心肌梗死组(MI),心肌梗死运动组(ME),每组10只。采用冠状动脉左前降支结扎手术建立心肌梗死模型。sham组除不进行结扎外其余手术过程同其他组。ME组心梗手术1周后进行为期8周间歇有氧运动。实验结束后,采用二维超声心动图检测大鼠心室厚度和射血功能。称量计算心重/体重比值(HW/BW)、左心室重量/体重比值(LVW/BW)。采用ELISA检测血浆和心肌组织胰岛素样生长因子-1(IGF-1)的表达水平。采用RT-qPCR检测心肌组织GATA4和Nkx 2.5mRNA水平,衡量心肌细胞的增殖能力。采用ELISA检测心肌端粒酶活性,并采用RT-qPCR检测心肌组织端粒酶逆转录酶(TERT)和自身携带的RNA(TERC)的mRNA水平。研究结果:1.心梗手术使大鼠心脏心室壁变薄,射血功能降低。与sham组相比,MI组射血分数(EF)明显降低(P≤0.01),HW/BW 和 LVW/BW 也明显降低(P≤0.05vs.P≤0.05);8 周间歇有氧运动后,ME组心脏舒张末期左室后壁厚度与舒张末期室间隔厚度之和(LVPWd+IVSd),EF 值均比 MI 组明显增高(P≤0.05 vs.P ≤0.05),HW/BW 和 LVW/BW也有明显升高(P≤ 0.05 vs.P≤ 0.05)。2.间歇有氧运动使心梗后大鼠心肌细胞增殖程度被激活。ME组血浆和心肌组织内IGF-1的水平比MI组明显升高(P≤0.05vs.P≤0.05)。与sham组相比,MI组心肌Nkx 2.5的mRNA表达水平明显升高(P≤ 0.05);而ME组心肌GATA4和Nkx 2.5 mRNA表达水平比MI组显著升高(P≤0.01 vs.P≤0.05)。3.间歇有氧运动能明显升高心梗大鼠心肌组织的端粒酶表达水平。有氧运动使ME组心肌组织端粒酶水平与MI组相比显著升高(P ≤ 0.01)。与Sham组相比,MI组心肌TERT mRNA表达水平明显增高(P ≤ 0.05);而ME组心肌TERC和TERT的mRNA表达水平都比MI组显著增高(P≤0.01vs.P≤0.01)。研究结论:1.间歇有氧运动能明显促进MI后大鼠心肌细胞增殖,改善心功能。2.间歇有氧运动能激活MI后大鼠心肌细胞端粒酶活性,进而促进心肌细胞增殖。
[Abstract]:Objective: to investigate the effects of aerobic exercise on telomerase activity and cardiomyocyte proliferation after myocardial infarction (MI) in rats. Randomly divided into sham operation group, myocardial infarction group, myocardial infarction group, myocardial infarction exercise group, 10 rats in each group. Coronary artery left anterior descending branch ligation operation was used to establish myocardial infarction model. Sham group except for no ligation of the rest of the operation process with other groups. ME. Group A received intermittent aerobic exercise for 8 weeks after myocardial infarction operation for 1 week. The ventricular thickness and ejection function of rats were measured by two-dimensional echocardiography. The ratio of heart weight to body weight was calculated and the ratio of left ventricular weight to body weight was measured. The expression of insulin-like growth factor-1 IGF-1 in plasma and myocardium was detected by ELISA. The levels of GATA4 and Nkx 2.5mRNA in myocardial tissue were detected by RT-qPCR.
