持续与间歇跑台运动对大鼠血清及不同组织自由基代谢及抗氧化能力的比较研究

【摘要】:研究目的:探讨6周持续与间歇跑台运动对大鼠血清及不同组织(心肌,肝脏,股四头肌,腓肠肌,比目鱼肌)MDA含量变化及SOD、T-AOC活性变化的影响。研究方法:雄性SD大鼠24只,随机分为安静对照组(S组)、持续训练组(C组)和长时间歇训练组(I组),共3组,每组8只。运动组先进行为期1周的适应性跑台。适应性跑台训练按照以下3级负荷进行:(1)第1-2天以15m/min×20min;(2)第3-4天18m/min×20min;(3)第5-6天21m/min×20min。正式训练时,安静组不运动,正常饲养,持续6周。运动组第一周以22m/min的速度开始进行跑台训练,以后每周以1m/min的速度递增,直至第6周27m/min为止,6天/周。其中持续训练组每天训练一次,训练40min;长时间歇组每天训练2组,每组训练20min,组间休息3h。6周训练结束后不限饮水,禁食24h,处死分别取出心尖血5ml,心肌,肝脏,股四头肌,比目鱼肌,腓肠肌等组织置于-80℃冰箱待测,全程在冰上进行,最后采用比色法(试剂盒)测定大鼠的MDA、SOD、T-AOC等指标。研究结果:1.MDA变化:6周持续与间歇跑台运动后,持续运动组血清MDA含量为5.7±0.49nmol/ml,与安静对照组相比显著降低(P0.05),长时间歇运动组血清MDA含量为3.65±0.68nmol/ml,与安静组相比有着极显著的差异(P0.01),且长时间歇组与持续训练组相比呈现出了显著差异(P0.05);心肌MDA含量持续训练组与安静对照组相比无明显差异(P0.05),长时间歇组与安静对照组相比,差异显著(P0.05),长时间歇组与持续组相比,显著下降(P0.05);肝脏MDA含量长时间歇组与安静对照组相比明显降低(P0.01),长时间歇组与持续训练组相比差异极显著(P0.01);股四头肌和腓肠肌MDA含量较安静组相比,有下降的趋势,但差异性分析无统计学意义(P0.05)而比目鱼肌MDA含量持续组与对照组相比显著降低(P0.05),长时间歇组与对照组相比降低的趋势极显著(P0.01),且长时间歇组与持续组相比也呈现出显著下降趋势(P0.05)。2.SOD变化:运动组大鼠血清SOD的数值有上升的趋势,持续组与对照组相比差异性不显著(P0.05)长时间歇组与对照组和持续组相比差异极显著(P0.01)。心肌SOD值运动组与安静组相比有升高的趋势,且长时间歇组与对照组相比有极显著差异(P0.01),与持续组相比,也呈现出显著差异(P0.05)。肝脏SOD值两运动组与安静对照组相比,有显著性的差异(P0.01);长时间歇组与持续组相比差异极其显著(P0.01)。股四头肌和腓肠肌SOD含量两运动组与对照组相比呈现上升趋势,但无统计学差异(P0.05),而比目鱼肌SOD含量长时间歇组与对照组和持续组相比差异显著(P0.05)。3.T-AOC变化:持续组和长时间歇组血清T-AOC活性与安静组相比明显升高,且差异性显著(P0.05和P0.01);心肌T-AOC活性长时间歇组与安静对照组和持续组相比有显著差异(P0.01和P0.05),持续组与对照组相比无统计学差异(P0.05);肝脏T-AOC活性两运动组与安静组相比,活性明显增强,具有极显著的差异(P0.01),且长时间歇组与持续组相比也具有显著性差异(P0.05);股四头肌和腓肠肌T-AOC活性与安静组相比有所升高,但没有显著性差异(P0.05);比目鱼肌T-AOC活性运动组与安静组相比有明显增强的趋势(P0.05),持续组与长时间歇组相比无统计学意义(P0.05)。研究结论:(1)经过6周持续与间歇跑台训练后,两种不同运动方式的大鼠体重增长幅度显著低于安静对照组,且长时间歇组显著低于持续组,推测中等强度的长时间歇有氧运动有助于体重控制。(2)经过6周持续与间歇跑台运动训练后,持续组与长时间歇组血清,心肌,肝脏和骨骼肌自由基MDA的含量较对照组呈现下降趋势,两运动组SOD和T-AOC的活性较安静组均呈现增强趋势,提示适宜的运动有助于提高机体的抗氧化能力。(3)经过6周持续与间歇跑台训练后,长时间歇训练组比目鱼肌SOD、T-AOC的活性显著高于股四头肌和腓肠肌,推测骨骼肌自由基的代谢可能与肌纤维类型有关。(4)在运动量相同的前提下,经过6周持续与间歇跑台训练后,长时间歇训练组血清和心肌自由基的代谢显著优于持续组和对照组,提示长时间歇训练组更利于自由基代谢。(5)在运动量相同的前提下,经过6周持续与间歇跑台训练后,长时间歇训练组肝脏自由基的代谢显著优于持续组和对照组,提示长时间歇训练有利于提高机体肝脏的解毒能力。
