总磷总氮测定仪的研制与优化

【摘要】:总磷总氮是衡量海洋生态环境的重要指标,若海洋水体中总磷总氮浓度过高,则会造成水体富营养化等生态问题,准确测定总磷总氮含量对于海水富营养化预警及保护海洋生态环境意义重大。海水中总磷总氮的国标检测方法操作繁琐、分析时间长,不能满足海水中总磷总氮的连续在线监测要求,因此,研发一套在线快速检测海水中总磷总氮的自动测定仪,对于提高我国海水水质监测水平具有重要意义。本研究采用微波-过硫酸盐消解技术结合顺序注射-阀上实验室技术测定水样中总磷总氮,并对消解条件、总磷显色条件及总氮还原、显色条件进行了优化。干扰离子实验和盐度干扰实验表明,海水中主要成分离子和盐度对本方法测定会产生干扰,可通过配制相应盐度的系列标准溶液来消除干扰。在最优消解、测定条件下,总磷总氮均在0~5 mg·L~(-1)范围内有良好线性,线性相关系数均为0.999,总磷精密度2.47%,总氮精密度2.39%,总磷检出限0.0050 mg·L~(-1),总氮检出限0.0066 mg·L~(-1),总磷加标回收率大于94%,总氮加标回收率大于96%,均符合海水中总磷总氮测定要求。在前期实验数据基础上,设计完成了总磷总氮自动测定装置,提出了装置的系统结构、各部分硬件、装置系统流程及仪器整体外观设计图,为总磷总氮测定仪的研制打下了基础。
[Abstract]:Total phosphorus nitrogen is an important index to measure the marine ecological environment. If the concentration of total phosphorus nitrogen in marine water is too high, it will lead to eutrophication of water body and other ecological problems. Accurate determination of total phosphorus nitrogen content is of great significance for eutrophication early warning and protection of marine ecological environment. The national standard detection method of total phosphorus total nitrogen in seawater is cumbersome and the analysis time is long. It can not meet the requirement of continuous on-line monitoring of total phosphorus nitrogen in seawater. Therefore, a set of automatic instrument for on-line rapid detection of total total phosphorus nitrogen in sea water has been developed. It is of great significance to improve the monitoring level of sea water quality in China. In this study, the total phosphorus nitrogen in water samples was determined by microwave persulfate digestion technology combined with sequential injection-valve laboratory technology, and the digestion conditions were analyzed. The color reaction conditions of total phosphorus and total nitrogen were optimized. The interference ion experiment and salinity interference experiment showed that the main component ions and salinity in seawater would interfere with the determination of this method. The interference can be eliminated by preparing a series of standard solutions with corresponding salinity. Under the optimum digestion and the determination conditions, the total phosphorus nitrogen has a good linearity in the range of 0 ~ 5 mg 路L ~ (-1). The linear correlation coefficients were 0.999, the precision of total phosphorus was 2.47, the precision of total nitrogen was 2.39, the detection limit of total phosphorus was 0.0050 mg 路L ~ (-1), the detection limit of total nitrogen was 0.0066 mg 路L ~ (-1) ~ (-1), the recovery rate of total phosphorus was more than 944.The total nitrogen recovery was more than 96. Based on the previous experimental data, An automatic total phosphorus nitrogen measuring device is designed and completed. The system structure, the hardware of each part, the system flow and the overall design diagram of the instrument are put forward, which lays a foundation for the development of the total phosphorus total nitrogen measuring instrument.
