新型碳点的制备及在分析检测方面的应用研究

【摘要】:近年来,碳量子点(又名碳点、碳纳米点、C-dots、CDs)因为其优异的荧光特性而受到了大家的广泛关注。由于碳量子点还具有尺寸小、分布窄、毒性低和生物相容性好等特点,被广泛应用于各种生物成像、生物传感、药物装载与运输以及疾病治疗等方面。本文就是利用了碳量子点优异的荧光特性和对一些物质灵敏的响应性,将其应用于生物分析检测、生物药物的运载等方面。主要工作内容如下:一、选取一种廉价易得的原料三聚氰酸,通过水热法合成了一种荧光性能良好的三聚氰酸碳点,并借助三聚氰胺和三聚氰酸之间强烈的三重氢键作用,通过三聚氰胺分子对碳量子点的荧光强度的影响来实现对三聚氰胺浓度的定量检测。二、以尿素和乙二胺为原料通过微波法合成一种表面充满氨基的碳量子点。又以聚天冬氨酸、聚乙二醇为原料,合成了一种含有一段离子嵌段的,由聚乙二醇(PEG)和聚天冬氨酸(PAsp)构成的二嵌段共聚物。然后通过静电作用自组装方式形成一种以碳量子点为内核,以二嵌段共聚物聚乙二醇聚天冬氨酸(PEG-b-PAsp)为外壳的具有良好荧光发射的荧光静电胶束。将静电胶束用于金属离子的检测,测试结果表明形成静电胶束后对金属离子的检测灵敏度明显增强。接着对静电胶束进行了药物运载等方面的研究,并通过细胞毒性测试、细胞内吞实验等证明了静电胶束的无毒性、良好的生物相容性,能够作为一种优良的纳米药物载体。
[Abstract]:In recent years, carbon quantum dots (also known as carbon, carbon nanotubes, C-dots, CDs) because of its excellent fluorescence properties and by all the attention. Because the carbon quantum dot has the features of small size, narrow distribution, low toxicity and good biocompatibility, is widely used in a variety of biological imaging, biological sensing, drug loading and transportation as well as disease treatment. This article is the use of the fluorescence properties of carbon quantum dots and some excellent material sensitive response, its application in biological analysis and detection, such as carrying biological drugs. The main work contents are as follows: first, choose a cheap raw materials melamine cyanate, cyanuric acid carbon a good fluorescence properties were synthesized by hydrothermal method, and with the help of between melamine and cyanuric acid, three strong hydrogen bonding interactions, affect the fluorescence intensity by melamine molecules on carbon quantum dots to To realize the quantitative detection of melamine concentration. With polyaspartic acid, polyethylene glycol as raw materials to synthesize a contains a ion block, by polyethylene glycol (PEG) and poly (aspartic acid PAsp) two block copolymer composition. Then through electrostatic self-assembly to form a carbon quantum dots as the core, with two polyethylene glycol copolymer of polyaspartic acid (PEG-b-PAsp) with fluorescence emission of good beam static rubber shell. The electrostatic detection of metal ions for micelles. The test results show that the formation of micelles after electrostatic detection sensitivity to metal ions significantly enhanced. Then study drug delivery and other aspects of the electrostatic micelles, and through cytotoxicity test, endocytosis experiments proved that electrostatic glue The nontoxic and good biocompatibility of the bundle can be used as a fine carrier for nanoscale drugs.
