唾液及头发中内源性大麻素的高效液相色谱-串联质谱检测

【摘要】:基于高效液相色谱-串联质谱仪建立了检测唾液和头发中花生四烯酸乙醇氨(AEA)和2-花生四烯酸甘油(2-AG)含量的方法,并用于检测157名在校大学生唾液和头发中的AEA和2-AG,探讨AEA和2-AG对性别和肥胖的影响.唾液中的待测物采用乙酸乙酯液液萃取提纯;头发中的待测物经甲醇孵育,用固相萃取柱萃取提纯.在大气压化学电离正离子模式和多反应监测方式下进行定性测试,以外标法进行定量测试.方法验证结果显示,唾液AEA,2-AG检测限分别为0.07和0.40 ng/m L;头发AEA,2-AG检测限分别为0.7和3.2 pg/mg;日内、日间变异系数均小于15%,回收率为97.9%~108.0%.人群研究结果表明,肥胖者唾液和头发AEA,2-AG含量均显著高于非肥胖者(p0.010);男性唾液2-AG含量显著低于女性(p0.050),但AEA含量没有性别差异(p=0.844);男性头发2-AG含量显著高于女性(p0.010),但AEA含量没有性别差异(p=0.456).
[Abstract]:Based on high performance liquid chromatography-tandem mass spectrometer, a method for the determination of arachidonic acid ethanolamine (AEAA) and 2-arachidonic acid glycerin (2-AGG) in saliva and hair was established.AEA and 2-AGG in saliva and hair of 157 college students were detected to investigate the effects of AEA and 2-AG on gender and obesity.The samples in saliva were extracted with ethyl acetate, and those in hair were incubated with methanol and extracted by solid phase extraction column.The qualitative test was carried out under the atmosphere pressure chemical ionization positive ion mode and the multi-reaction monitoring mode, and the quantitative test was carried out by external standard method.The results showed that the detection limit of AEAA 2-AG in saliva was 0.07 and 0.40 ng/m / L, the detection limit of AEAA 2-AG in hair was 0.7 and 3.2 PG / mg, and the coefficient of variation in day was less than 15 and the recovery rate was 108.0%.The results of the population study show thatThe content of AEAA 2-AG in saliva and hair of obese group was significantly higher than that in non-obese group (p0.010), the content of 2-AG in male saliva was significantly lower than that in female (P 0.050), but the content of AEA was not different in sex. The content of 2-AG in male hair was significantly higher than that in female (P 0.010 0), but the content of AEA in male was not different from that in female (P 0.456).
