驾驶员血清5-HT、5-HTP及BDNF水平与事故倾向性的相关研究

【摘要】:目的通过广西驾驶员血清中5-羟色胺(5-hydroxytryptamine,5-HT)、5-羟色氨酸(5-hydroxytryptophan,5-HTP)及其脑源性神经营养因子(brain-derived neurotrophic factor, BDNF)含量的测定,寻求事故倾向性驾驶员的生物标志物,探讨事故倾向性驾驶员5-HT、5-HTP、BDNF水平与其发生事故的相关因素是否存在关联。从生物学角度初步探索事故倾向性产生的原因和作用机制,为今后驾驶员的筛检进而减少交通事故的发生提供科学依据。方法采用病例对照的研究方法,通过调查问卷,根据研究对象的纳入和排除标准选取2014年广西事故倾向性驾驶员139例和正常对照驾驶员127例。通过高效液相色谱法(High performance liquid chromatography, HPLC)测定166例研究对象(病例组89例,对照组77例)血清中5-HT、 5-HTP水平。采用酶联免疫吸附试验(enzyme linked immunosorbent assay, ELISA)测定100例研究对象(病例组和对照组各50例)血清BDNF水平。应用EpiData3.1和SPSS16.0统计软件进行数据的录入和统计分析。采用t检验、χ2检验、Pearson相关分析、非条件Logistic回归分析的统计方法。结果 (1)病例组血清中5-HT (125.45±66.41)μg/L和5-HTP (225.06±55.54) μg/L均低于对照组5-HT (146.29±62.33) μg/L和5-HTP (249.33±58.16) μg/L (P0.05); 5-HT/5-HTP比值两组间差异无统计学意义。(2)5-HTP与5-HT呈正相关趋势(相关系数r=0.784,P0.05);病例组中5-HTP与5-HT呈正相关趋势(相关系数r--0.743,P0.05);对照组中5-HTP与5-HT呈正相关趋势(相关系数r=0.813,P0.05)。(3)驾龄至10年的驾驶员中,病例组5-HT(123.13±48.38)μg/L和5-HTP(219.65±45.98)μg/L均低于对照组5-HT(154.01±66.56)μg/L和5-HTP(254.00±46.85)μg/L(P0.05).(4)病例组中汉族驾驶员5-HTP(212.61±37.51)μg/L低于对照组中汉族驾驶员(246.37±44.39)μg/L(P0.05);病例组中其他少数民族驾驶员5-HT(113.14±54.13)μg/L低于对照组中其他少数民族驾驶员(156.53±60.68)μg/L(P0.05);病例组中汉族驾驶员5-HTP(212.61±37.51)μg/L低于病例组中其他少数民族驾驶员(241.98+70.83)μg/L(P0.05)。(5)非条件Logistic回归结果显示5-HIT、5-HTP和锻炼频率与事故倾向性有关,可能为其保护因素。(6)病例组血清BDNF水平(3.18±2.19)ng/mL高于对照组水平(2.23±1.43)ng/mL(P0.05);病例组驾龄10年的驾驶员血清BDNF水平高于对照组(P0.05);病例组初中及以下的驾驶员血清BDNF水平高于对照组(P0.05);病例组其他民族的驾驶员血清BDNF水平高于对照组(P0.05)。(7)非条件Logistic回归结果显示睡眠时间和锻炼频率与事故倾向性有关,可能为其保护因素。结论血清5-HT、5.HTP、BDNF水平与事故倾向性驾驶员发生事故有关联。血清5-HTP与5-HT呈正相关趋势,两组血清5-HT/5-HTP比值没有明显差异,提示体内血清5-HTP与5-HT的浓度比例变化一致。血清5-HT、 5-HTP、睡眠时间和锻炼频率可能为事故倾向性保护因素。
[Abstract]:Objective to explore the biomarkers of accident prone drivers by the determination of serotonin 5-hydroxytryptamine 5-hydroxytryptophanine 5-HTP (5-hydroxytryptophanine 5-HTP) and brain-derived neurotrophic factor (brain-derived neurotrophic factor, BDNF) in Guangxi drivers. To investigate the correlation between BDNF level and accident prone drivers. From the biological point of view, the causes and mechanism of accident tendency are preliminarily explored, which provides scientific basis for driver screening and reducing the occurrence of traffic accidents in the future. Methods 139 accident prone drivers in Guangxi in 2014 and 127 normal control drivers were selected according to the inclusion and exclusion criteria of the subjects.
