【摘要】:催乳素是在泌乳启动和维持中起重要作用的激素。催乳素可以促进乳腺导管延伸、乳汁生成,促进乳成分的合成,尤其影响乳脂的合成。为了研究催乳素对乳脂合成的调控机制,本研究使用多巴胺(D2)受体激动剂喹高利特抑制泌乳大鼠催乳素的释放,采用数字基因表达谱分析比对喹高利特处理组(Quinagolide)与对照组(Control)大鼠泌乳乳腺乳脂合成基因的差异表达情况。为了验证测序结果的准确性,本研究选取了10个基因,使用荧光定量PCR技术测定了其表达情况。本试验使用转录组测序的方法构建了两组(Quinagolide、Control)大鼠乳腺的总RNA文库,平均产生14,458,067条clean reads。两个文库分别得到12843和13639个基因。以|log2Ratio(Quinagolide/Control)|≥2,FDR≤0.001为基因显著差异标准,分析两组乳腺组织基因差异表达情况。对照组与喹高利特处理组比对,显著差异表达的基因共有2823个,其中下调基因262个,上调基因2561个,分别占到差异表达基因总数的3.9%和42.3%。转录水平的研究结果表明:(1)喹高利特抑制催乳素主要影响乳腺上皮细胞分化、生长,进而影响乳产量。(2)催乳素抑制直接调节脂肪酸结合蛋白(FABP3)、乙酰辅酶A羧化酶(ACC)、脂肪酸脱氢酶(FADS1)、胰岛素诱导基因1(INSIG1)的转录水平。
[Abstract]:Prolactin is a hormone that plays an important role in the initiation and maintenance of lactation. Prolactin can promote breast duct extension, milk production and the synthesis of milk components. In order to study the regulation mechanism of prolactin on the synthesis of milk fat, we used the dopamine D _ 2) receptor agonist quinolide to inhibit the prolactin release in lactating rats. Quinagolide (Quinagolide) and control group (control) were compared by digital gene expression profile analysis. Differential expression of milk fat synthesis gene in rat lactation mammary gland in order to verify the accuracy of sequencing results. In this study, 10 genes were selected and their expression was determined by fluorescence quantitative PCR. Two groups of Quinagolide were constructed by transcriptome sequencing.
