高原地区55型腺病毒的病原分离及全基因序列分析

【摘要】:目的探讨高原地区55型腺病毒(HAdV-B55)基因变异特点。方法采集驻西藏某部队55型腺病毒感染者咽拭子标本,采用HEp-2分离病毒株,通过分片段PCR扩增和测序获得HAdV-B55毒株的全基因序列,通过与内地毒株进行序列比对,发现其变异特点。结果从55型腺病毒感染者咽拭子标本分离得到HAdV-B55毒株,命名为LS89/Tibet/2016,通过PCR产物测序拼接,获得了本次流行的HAdV-B55毒株的全基因序列,Gen Bank登录号为KY002683。通过MEGA6软件比对,发现该病毒基因序列并未发生大片段重组,与内地以往流行毒株(QS-DLL株)序列相似性为99.9%。结论本次分离出的病毒基因序列未发生大片段重组。本研究为揭示55型腺病毒的进化特点及今后高原地区制订疾病防控策略提供了依据。
[Abstract]:Objective to investigate the variation characteristics of adenovirus 55 (HAdV-B55) gene in plateau area. Methods the throat swabs of adenovirus type 55 infection in a military unit in Tibet were collected and the virus strains were isolated by HEp-2. The whole gene sequence of HAdV-B55 strain was obtained by fragment PCR amplification and sequencing. Results HAdV-B55 strain was isolated from throat swabs of adenovirus type 55 infection and named LS89/Tibet/2016. The whole gene sequence of HAdV-B55 strain was obtained by sequencing and splicing the PCR product. The accession number of Genin Bank was KY002683. The result was compared by MEGA6 software. It was found that there was no large fragment recombination in the gene sequence of the virus. The QS-DLL strain and the previous prevalent virus strain in mainland China. The sequence similarity is 99.9. Conclusion there is no large fragment recombination of the virus gene sequence isolated in this paper. This study provides a method to reveal the evolutionary characteristics of adenovirus type 55 and to formulate disease prevention and control strategies in the plateau region in the future. Basis
