健康促进生活方式和烟碱型乙酰胆碱受体亚单位a5基因多态与肺癌发病风险的病例对照研究

【摘要】：目的:研究健康促进生活方式和烟碱型乙酰胆碱受体亚单位a5(CHRNA5)-rs 17486278位点基因多态与肺癌发病风险的关系。方法:以2014年12月至2015年12月唐山市工人医院确诊的216肺癌例患者为病例组,按照年龄、性别、居住地等配对条件选取639例健康体检人群为对照组。采用聚合酶链反应-限制性片段长度多态性方法(PCR-RFLP)检测CHRNA5-rs 17486278位点基因多态,结合两组人群的健康资料,分析健康促进生活方式和遗传因素与肺癌发病风险的关系。结果:Logistic回归显示健康促进生活方式六个维度中的压力管理与肺癌发生有关,OR值为1.770(95%CI:1.262~2.483,P=0.001);病例组和对照组间CHRNA5-rs 17486278等位基因分布频率之间比较,差异有统计学意义(X~2=10.067,P=0.002);压力管理与吸烟指数、吸烟指数与CHRNA5-rs 17486278位点基因型对肺癌发生的交互作用,差异有统计学意义(X~2交互作用=4.86,P交互作用=0.034;X~2交互作用=8.62,P交互作用=0.003)。结论:健康促进生活方式和CHRNA5-rs 17486278位点基因多态存在协同作用,增加肺癌发生的风险。
[Abstract]:Objective: to study the relationship between the health promoting lifestyle and the polymorphism of A5CHRNA5- rs 17486278 gene in the nicotinic acetylcholine receptor subunit. Methods: from December 2014 to December 2015, 216 lungs were diagnosed in Tangshan workers' hospital. Cancer patients as case group, CHRNA5-rs 17486278 locus polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method, and the health data of the two groups were analyzed according to the matched conditions of age, sex and residence. The relationship between health promotion lifestyle and genetic factors and lung cancer risk was analyzed. Results: the OR value of stress management in six dimensions of health promotion lifestyle was 1.77095 CI: 1.2622.483P 0.001; case group and control group. The frequency of CHRNA5-rs 17486278 allele distribution was compared. The difference was statistically significant (P < 0.05). The interaction between stress management and smoking index, smoking index and genotype of CHRNA5-rs 17486278 locus on the occurrence of lung cancer. The difference was statistically significant (P < 0.05). Conclusion: there is a synergistic effect between health promoting lifestyle and polymorphism of CHRNA5-rs 17486278 locus gene, which increases the risk of lung cancer.
