甘蓝型油菜BnGPAT9基因表达模式及其苗期非生物胁迫表达分析

【摘要】:为揭示油菜BnGPAT9基因的功能及其在非生物胁迫中的表达变化,克隆甘蓝型油菜BnGPAT9基因(GenBank:KX344511.1)并分析表达模式。发现该基因全长1 131bp,编码376个氨基酸残基。序列比对及系统进化树分析表明BnGPAT9与其它植物的GPAT9有很大的相似性,在生物进化过程中具有很高的保守性。实时荧光定量PCR分析表明,BnGPAT9基因在甘蓝型油菜中的表达具有组织特异性,在开花后1~4周幼嫩的种子中表达较强,而在茎、花、叶、根和角果皮中表达较弱;5~6片真叶期盆栽苗的非生物胁迫表达分析显示,短期干旱和高温能够诱导和促进叶片BnGPAT9基因的表达;低温、6-BA、ABA和盐胁迫抑制其表达;水渍对BnGPAT9基因的表达影响呈"Z"字形,12h出现最低值,但24h后又开始回升。
[Abstract]:In order to reveal the function of BnGPAT9 gene and its expression changes in abiotic stress in rapeseed, BnGPAT9 gene of Brassica napus L. was cloned into GenBank: KX344511.1) and its expression pattern was analyzed. The total length of the gene was 1131 BP, encoding 376 amino acid residues. Sequence alignment and phylogenetic tree analysis showed that BnGPAT9 was similar to GPAT9 of other plants. The real-time fluorescence quantitative PCR analysis showed that the expression of BnGPAT9 gene was tissue specific in Brassica napus, and was strongly expressed in young seeds 1 week after anthesis, but in stems, flowers and leaves. Abiotic stress analysis showed that short-term drought and high temperature could induce and promote the expression of BnGPAT9 gene in leaves, but low temperature and salt stress inhibited the expression of BnGPAT9 gene. The effect of water on the expression of BnGPAT9 gene was the lowest in 12 h, but it began to rise again after 24 hours.
