不同筋度小麦及其近缘种在Glu-3位点上LMW-GS基因单元型的分离和鉴定

【摘要】:低分子量麦谷蛋白约占小麦种子贮藏蛋白的三分之一,对面团延展性和食品加工品质有着重要影响。普通小麦及其近缘种是小麦遗传改良的重要基因资源。本研究利用小麦Glu-A3和Glu-B3基因的特异引物对强筋型、中筋型和弱筋型小麦以及小麦近缘种共计21份材料的LMW-GS基因进行扩增和鉴定,共计发现了33个新的核苷酸变异类型和26个肽链变异类型,包括在GluA-3位点上得到19个GluA-3基因变异类型和19个对应的肽链变异类型,在GluB-3位点上得到5个Glu-B3基因变异类型和3个肽链变异类型,在GluD-3位点上得到9个Glu-D3基因变异类型和4个肽链变异类型。此外,还发现3个半胱氨酸数目特殊的亚基类型,分别是来自品种师栾02-1和栽培二粒小麦DM2含有9个半胱氨酸残基的GluA3-18基因、野生一粒小麦B03含有9个半胱氨酸残基的GluA3-312基因以及偃展4110含有7个半胱氨酸残的GluD3-13基因。3个半胱氨酸数目特殊的亚基类型,可能属于优良品质亚基。对普通小麦及其近缘种中这些Glu-3位点等位变异的鉴别,进一步完善了小麦低分子量麦谷蛋白亚基的构成,并为小麦品质育种中亲本的选择提供了相应依据。
[Abstract]:Low molecular weight glutenin accounts for about 1/3 of wheat seed storage protein. The ductility and food processing quality of the opposite group have an important effect on wheat. The common wheat and its relative species are important genetic resources for wheat genetic improvement. In this study, the strong gluten type was identified by using the specific primers of Glu-A3 and Glu-B3 genes in wheat. The LMW-GS gene of 21 accessions of middle gluten type and weak gluten type wheat and their relative species were amplified and identified. A total of 33 new nucleotide variation types and 26 peptide chain variant types were found. 19 GluA-3 gene variants and 19 corresponding peptide chain variants were obtained at GluA-3 loci, and 5 Glu-B3 gene variants and 3 peptide chain variant types were obtained at GluB-3 loci. Nine Glu-D3 gene variants and four peptide chain variants were obtained at GluD-3 loci. In addition, three specific subunit types of cysteine were found. The GluA3-18 gene from the cultivar Luan Luan 02-1 and the cultivated wheat DM2 contains 9 cysteine residues, respectively. Wild wheat B03 contains 9 cysteine residues of GluA3-312 gene and Yanchan 4110 contains 7 cysteine residues of GluD3-13 gene. The allelic variation of these Glu-3 loci in common wheat and its relative species was identified, which further improved the composition of wheat low molecular weight glutenin subunits. It also provides the basis for the selection of parents in wheat quality breeding.
