牦牛DGAT2基因intron5和intron6多态性及其对乳品质性状的影响

【摘要】:乳脂是乳的重要组成部分,高乳脂率是牦牛(Bos grunniens)乳的主要特征。二酰基甘油酰基转移酶2(diacylglycerol acyltransferase 2,DGAT2)调控乳中甘油三酯(triacylglyceride,TRIG)合成,是乳脂肪性状的重要候选基因。本实验应用聚合酶链式反应-单链构象多态性(polymerase chain reaction-single strand conformation polymorphism,PCR-SSCP)技术检测甘南牦牛、天祝白牦牛及大通牦牛DGAT2基因多态性,并分析基因突变对甘南牦牛乳品质性状影响。结果表明,三类群牦牛在DGAT2基因intron5区检测到A_1、B_1、C_1 3种等位基因,存在c.613-214 AG和c.613-337 CA的SNPs;intron6区检测到A_2、B_2、C_2 3种等位基因,存在c.787+616 GA和c.787+691 TG的SNPs;三类群牦牛DGAT2基因两区域均表现为中度多态,4个突变位点在群体中形成7种单倍型,各位点间存在连锁关系且接近于连锁平衡。甘南牦牛DGAT2基因intron5区突变影响乳脂率,第2胎携带等位基因B_1的个体乳脂率显著高于未携带个体(P0.05);intron6区突变影响乳脂率、乳蛋白率及总固体物质含量,携带等位基因A_2的个体乳脂率、总固体物质含量较高且第2胎母牦牛显著高于未携带者(P0.05),即选留携带等位基因A的个体有增加后代群体乳脂率及总固体物质含量的趋势;第2胎母牦牛基因型A_2C_2个体乳蛋白率最高,且A_2C_2、B_2B_2型个体显著高于其他基因型(P0.05);单倍型H1H3个体的乳品质性状也高于其他单倍型。DGAT2基因intron5和intron6区突变可作为甘南牦牛乳品质性状潜在的遗传标记。本研究为牦牛乳品质性状候选基因分子遗传研究积累数据。
[Abstract]:Milk fat is an important part of milk, and high milk fat rate is the main characteristic of Bos grunniens) milk. Diacylglycerol acyltransferase 2 (DGAT2) regulates the synthesis of triacylglyceride triacylglyceride triIGl in milk. Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCPP-PCR) was used to detect DGAT2 gene polymorphism in Gannan yak, Tianzhu white yak and Datong yak. The effect of gene mutation on milk quality of Gannan yak was analyzed. The results showed that three groups of yaks had detected A1B1C 13 alleles in the intron5 region of DGAT2 gene, and there were 23 alleles in A2B2C2CC6 region of c.613-214 AG and c.613-337 CA, respectively, in the SNPs intron 6 region of C. 613-214 AG and c.613-337 CA. SNPs of c. 787,616 GA and c. 787 691 TG existed, and the two regions of DGAT2 gene were moderately polymorphic, and the four mutation loci formed 7 haplotypes in the populations. There was a linkage relationship and close to linkage equilibrium. The mutation of intron5 region of DGAT2 gene in Gannan Yak affected the milk fat rate. The milk fat rate of individuals carrying allele B-1 in the second embryo was significantly higher than that in the non-carrying individuals (P 0.05 intron 6), and the milk fat rate was affected by the mutation of the intron5 region of the DGAT2 gene in Gannan Yak. Milk protein rate and total solid matter content, individual milk fat rate with allelic A _ 2, The content of total solid matter in the second female yak was significantly higher than that in the non-carrier group P0.05, that is, the individuals with allele A had a tendency to increase the milk fat rate and the total solid matter content of the progeny population. The milk protein rate of the second female yak genotype ACTH 2C2 was the highest. The milk quality traits of the population with A2C2B2 were significantly higher than those of other genotypes P0.05. the milk quality traits of haplotype H1 H3 individuals were also higher than those of other haplotypes. DGAT2 gene intron5 and intron6 region mutations could be used as potential genetic markers for milk quality traits of Gannan yaks. This study is a potential genetic marker for milk quality traits of Gannan yaks. Molecular genetic study on candidate genes for Milk quality traits.
