【摘要】:目的:通过检测维吾尔族饲鸽者肺外周血Notch4基因甲基化程度探讨其临床意义。方法:根据纳入及排除标准连续收集2015年-2016年在新疆喀什病例资料及血样标本(总纳入60例,其中20例确诊维吾尔族饲鸽者肺患者为病例组,同期20例维吾尔族饲养鸽子未发病者为阴性对照组,同期20例维吾尔族未饲养鸽子健康体检者为正常对照组)。对所收集血样标本进行常规提取DNA,亚硫酸氢盐转化,PCR扩增,体外转录和RNase A特异性酶切,并行基质辅助激光解吸附电离飞行时间质谱分析技术检测Notch4基因甲基化情况。结果:检测Notch4片段的CpG位点总数6个(CpG_1,CpG_2/3,CpG_4,CpG_5,CpG_6),CpG_1,CpG_4,CpG_5,CpG_6在三组样本中的甲基化率分布差异无统计学意义(p0.05)。CpG_2/3在三组样本甲基化率分布差异具有统计学意义(p0.05)。病例组CpG_2/3甲基化率高于阴性对照组,差异有统计学意义(P0.05),病例组CpG_2/3甲基化率高于正常对照组,差异有统计学意义(P0.05),但阴性对照组与正常对照组CpG-2/3甲基化率差异无统计学意义(P0.05)。结论:Notch4基因甲基化通过炎症反应及信号通路调控肺纤维的发生发展。
[Abstract]:Objective: to investigate the clinical significance of Notch4 gene methylation in peripheral blood of pigeon feeding pigeons in Uygur nationality. Methods: according to the criteria of inclusion and exclusion, the data of Kashi cases and blood samples from 2015 to 2016 were collected. Among them, 20 cases were diagnosed as case group and 20 cases as negative control group. In the same period, 20 Uygur people without pigeon health examination were used as the normal control group. The blood samples were routinely extracted and amplified by hydrogen sulfite transformation PCR, transcribed in vitro and digested with RNase A specific enzyme. Simultaneous matrix assisted laser desorption ionization time of flight mass spectrometry was used to detect Notch4 gene methylation. Results: there was no significant difference in the distribution of methylation rate of Notch4 gene in the three groups by detecting the total number of CpG sites in Notch4 fragments, six CpG1 / CpG1 / CpG1 / CpG1 / CpG4 / CpG4 / CpG5 / CpG5 / CpG5 / CpG5CpG6 in all three groups of samples, and the number of CpG1CpG5CpG6 in the three groups was not significantly different from that of CpG1CpG5CpG6. There was significant difference in the distribution of methylation rate among the three groups. The CpG_2/3 methylation rate in the case group was higher than that in the negative control group. The difference was statistically significant (P 0.05). The rate of CpG_2/3 methylation in the case group was higher than that in the normal control group. The difference was statistically significant (P 0.05), but there was no significant difference in CpG-2/3 methylation rate between the negative control group and the normal control group. Conclusion the methylation of the 1: Notch4 gene regulates the development of lung fiber through inflammatory reaction and signal pathway.
