甘蓝型油菜DOF蛋白基因的克隆与生物信息学分析

【摘要】:Dof转录因子是植物特有的一类转录因子,含有一个独特的单锌指结构域,在植物的生长发育中起重要的调控作用。以甘蓝型油菜为试材,采用电子克隆和RT-PCR技术,以期克隆一个具有Zf-Dof结构域的蛋白基因cDNA序列。结果表明:该基因含有1284bp的开放阅读框,编码427个氨基酸,蛋白分子量为46.9kDa,等电点为8.86,为亲水性蛋白。经序列比对分析表明其与拟南芥CDF3基因有较高同源性,可能具有相似的功能。
[Abstract]:Dof transcription factors are plant specific transcription factors, which contain a unique single zinc finger domain and play an important role in plant growth and development. Using Brassica napus as a test material, electronic cloning and RT-PCR techniques were used. In order to clone the cDNA sequence of a protein gene with Zf-Dof domain, the results showed that the gene contained 1284bp open reading frame, encoding 427amino acids. The protein has a molecular weight of 46.9 kDa and an isoelectric point of 8.86, which is a hydrophilic protein. Sequence alignment analysis shows that the protein has high homology with Arabidopsis CDF3 gene and may have similar function.
