白介素6受体基因rs4129267位点多态性与汉族儿童哮喘易患性及血清IgE水平的关联研究

【摘要】：背景有研究证实,白介素6受体(IL-6R)基因中的rs4129267位点多态性与哮喘易患性显著相关,但尚无评估rs4129267位点多态性在汉族儿童中分布情况及其与血清IgE水平相关性的研究。目的研究IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性及血清IgE水平的关系。方法选取2013—2014年于广东省妇幼保健院儿科就诊的哮喘患儿410例为哮喘组,同期选取广东省妇幼保健院住院的非哮喘患儿513例为对照组。提取两组患儿基因组DNA,采用SNa Pshot法对IL-6R基因rs4129267、rs527236128位点进行基因分型,采用酶联免疫吸附试验(ELISA)法检测两组患儿血清总IgE水平。结果两组患儿rs4129267位点和rs527236128位点的基因型分布均符合Hardy-Weinberg平衡(χ~2=3.264、2.135,P0.05)。两组患儿rs4129267位点基因型频率和等位基因频率比较,差异有统计学意义(P0.05)。两组患儿rs527236128位点基因型频率和等位基因频率比较,差异无统计学意义(P0.05)。Logistic回归分析结果显示,TT基因型是儿童哮喘的影响因素(P0.05)。对照组患儿血清总IgE水平低于哮喘组(P0.05)。对照组、哮喘组组内不同基因型患儿血清总IgE水平比较,差异无统计学意义(P0.05);对照组各基因型患儿血清总IgE水平均低于哮喘组(P0.05)。结论 IL-6R基因rs4129267位点多态性与汉族儿童哮喘易患性有关,而与哮喘患儿血清IgE水平无关。
[Abstract]:Background rs4129267 polymorphism in the interleukin-6 receptor IL-6R) gene has been confirmed to be significantly associated with asthma susceptibility. However, the distribution of rs4129267 locus polymorphism in Han children and its correlation with serum IgE level have not been evaluated. Objective to study the relationship between IL-6R gene rs4129267 locus polymorphism and asthma susceptibility and serum IgE level in Han children. Methods 410 asthmatic children in Guangdong Maternal and Child Health Hospital from 2013 to 2014 were selected as asthma group. In the same period, 513 non-asthmatic children in Guangdong Maternal and Child Health Care Hospital were selected as control group. Genomic DNA was extracted from two groups of children, and IL-6R gene rs4129267rs527236128 was genotyped by SNa Pshot method. Serum total IgE levels were detected by Elisa. Results the genotype distribution of rs4129267 and rs527236128 loci in the two groups were in accordance with the Hardy-Weinberg equilibrium (蠂 2 / 264 / 2.135 P 0.05). The frequencies of rs4129267 loci and alleles were compared between the two groups. The frequency of genotype and allele of rs527236128 locus were compared between the two groups. The results of logistic regression analysis showed that the serum total IgE level in the control group was lower than that in the asthmatic group. The serum total IgE level in the asthmatic group was lower than that in the control group, and the serum total IgE level in the asthmatic group was higher than that in the control group. The serum total IgE level of the control group was lower than that of the asthmatic group (P 0.05). Conclusion the polymorphism of rs4129267 locus of IL-6R gene is related to the susceptibility to asthma in Han nationality children, but not to the serum IgE level of asthmatic children.
