盐和干旱胁迫下红麻HcWD40-1基因的克隆及表达特征

【摘要】:WD-重复蛋白(WD-repeat protein)含有WD-40基序,又称为WD40重复蛋白,在植物对非生物逆境胁迫响应过程中具有重要的调控作用。红麻(Hibiscus cannabinus)是重要的工业纺织原料,是盐碱地土壤改良的先锋作物。本研究以红麻转录组中与WD40基因高度相似的Unigene为参考,设计引物,进行反转录PCR扩增,经Sanger测序获得基因Hc WD40-1(Gen Bank登录号:KX711617)的c DNA序列,生物信息学分析表明,该基因开放阅读框为1 356 bp,编码451个氨基酸,具有7个典型的WD40结构域。q RT-PCR分析表明,Hc WD40-1基因受盐和干旱胁迫诱导表达,同时受脱落酸(abscisic acid,ABA)和茉莉酸甲酯(methyl jasmonate,Me JA)的诱导。研究结果表明,Hc WD40-1基因是ABA和Me JA信号转导途径、盐和干旱胁迫应答途径的关键枢纽基因。该研究结果为红麻耐盐、抗旱的脱落酸信号调控网络和茉莉酸信号调控网络的研究提供基础资料。
[Abstract]:WD- repeat protein contains WD-40 motif, also known as WD40 repeat protein, which plays an important role in plant response to abiotic stress. Hibiscus cannabinus is an important industrial textile material and a pioneer crop for soil improvement in saline-alkali soil. In this study, the highly similar Unigene gene of kenaf transcriptiongroup was used as reference, primers were designed and reverse transcription PCR was carried out. The c DNA sequence of the gene HC WD40-1(Gen Bank accession number: KX711617 was obtained by Sanger sequencing. Bioinformatics analysis showed that, The open reading frame of the gene was 1 356 BP, encoding 451 amino acids, with seven typical WD40 domains. Q RT-PCR analysis showed that the Hc WD40-1 gene was induced by salt and drought stress, and was also induced by Aba (abscisic acidida) and methyl jasmonate (me JAA). The results showed that the Hc WD40-1 gene was the key gene of ABA and me JA signal transduction pathway, and the key gene of salt and drought stress response pathway. The results provided the basic data for the study of salt tolerance, drought resistance abscisic acid signal control network and jasmonic acid signal control network.
