自噬相关基因微管相关蛋白1轻链3和叉头框蛋白O1在胃癌组织中的表达及其意义

【摘要】：目的:探讨自噬相关基因微管相关蛋白1轻链3(microtubule-associated protein 1 light 3,LC3)和叉头框蛋白O1(forkhead box protein O1,Fox O1)在胃癌组织中的表达及其意义。方法:应用免疫组织化学SP法检测29例胃癌组织和癌旁正常组织中自噬相关基因LC3和Fox O1的表达情况,分析自噬相关基因表达水平与临床病理特征之间的相关性。结果:免疫组化结果提示LC3蛋白、Fox O1蛋白在胃癌组织中的阳性表达率均高于癌旁正常组织(P0.01)。LC3蛋白表达水平与胃癌的分化程度、淋巴结转移、临床分期、侵及胃壁深度明显相关,与患者的性别、年龄、胃癌发生部位及大小无明显相关性;Fox O1蛋白表达水平与胃癌的分化程度明显相关,与患者的性别、年龄、胃癌发生部位、大小、淋巴结转移、临床分期、侵及胃壁深度无明显相关性。结论:自噬相关基因LC3和Fox O1在胃癌组织中表达水平增高,均与胃癌的分化程度相关,且Fox O1高表达是胃癌早期频发事件,提示自噬活动水平的上调对胃癌的发生、发展具有重要促进作用。
[Abstract]:Objective: to investigate the expression and significance of microtubule-associated protein 1 (3(microtubule-associated protein _ 1 light _ 3) and O1(forkhead box protein O _ (1) Fox O _ (1) in gastric carcinoma. Methods: immunohistochemical SP method was used to detect the expression of autophagy associated genes (LC3 and Fox O1) in 29 cases of gastric cancer and adjacent normal tissues, and the correlation between the expression level of autophagy related genes and clinicopathological features was analyzed. Results: the results of immunohistochemistry showed that the positive expression rate of LC3 protein in gastric carcinoma was higher than that in adjacent normal tissues. The expression level of LC3 protein was significantly correlated with the degree of differentiation, lymph node metastasis, clinical stage and depth of gastric wall invasion. There was no significant correlation between the expression of Fox O1 protein and sex, age, location and size of gastric cancer, but also with sex, age, location, size, lymph node metastasis, clinical stage of gastric cancer. There was no significant correlation between the depth of gastric wall invasion. Conclusion: the increased expression of autophagy related genes LC3 and Fox O1 in gastric carcinoma is related to the differentiation of gastric cancer, and the high expression of Fox O1 is a frequent event in the early stage of gastric cancer, suggesting that the upregulation of autophagy activity is associated with the occurrence of gastric cancer. Development plays an important role in promoting development.
