安肠汤对溃疡性结肠炎大鼠肠道真菌基因表达的影响

【摘要】:目的探讨安肠汤治疗溃疡性结肠炎的可能作用机制。方法将112只大鼠随机分为空白组、模型组、美沙拉嗪组、丽珠肠乐组和安肠汤高、中、低剂量组,每组16只。除空白组外其余各组大鼠建立溃疡性结肠炎模型,造模后第10天空白组和模型组大鼠每只以生理盐水3 ml灌胃;丽珠肠乐组以丽珠肠乐溶液(0.5亿活菌/ml)灌胃,每次0.12 ml;美沙拉嗪组以美沙拉嗪混悬液(5 mg/ml)灌胃,每次0.35 ml;安肠汤低、中、高剂量组分别给予安肠汤浓缩液1、5、10 ml/(kg·d),每日1次,共10天。取结肠病变肠段进行HE染色观察结肠黏膜组织形态学,取各组粪便标本应用实时荧光定量技术测定大鼠肠道5种真菌含量(包括白色念珠菌、曲霉菌、青霉菌、枝孢菌、隐球菌)。结果 HE染色结果显示,模型组大鼠结肠黏膜层几乎完全缺损,溃疡形成,黏膜充血糜烂,伴炎细胞浸润;各治疗组则较模型组出现改善。与空白组比较,其余各组大鼠肠道白色念珠菌、曲霉菌和青霉菌相对表达含量上明显升高,而枝孢菌和隐球菌相对表达含量降低(P0.05)。与模型组比较,各治疗组大鼠肠道白色念珠菌、曲霉菌和青霉菌相对表达含量明显降低,枝孢菌和隐球菌相对表达含量升高(P0.05)。安肠汤中剂量组在降低曲霉菌及升高枝孢菌、隐球菌相对表达含量方面优于安肠汤低、高剂量组,在降低白色念珠菌及青霉菌相对表达含量方面安肠汤高剂量组优于安肠汤低、中剂量组(P0.05)。结论安肠汤对溃疡性结肠炎大鼠肠道真菌具有明确的调节作用,可能是其治疗溃疡性结肠炎的作用机制之一。
[Abstract]:Objective to explore the possible mechanism of Anchang decoction in the treatment of ulcerative colitis. Methods 112 rats were randomly divided into blank group, model group, mesalazine group, Lizhu Changle group and Anchang decoction group with 16 rats in each group. The ulcerative colitis model was established in rats in each group except the blank group. After the establishment of the model, the rats in the 10th sky white group and the model group were given 3 ml normal saline, and the Lizhu Changle group was given Lizhu Changle solution (50 million live bacteria / ml), and the rats in the model group were given intragastric administration of Lizhu Changle solution (50 million live bacteria / ml). Each time 0.12 ml; the mesalazine group was perfused with 5 mg / ml of mesalazine suspension (0.35 ml / time); the low, medium and high dose group of Anchang decoction was given the concentrated solution of Anchang decoction for 10 days once a day. Colonic mucosal histomorphology was observed by HE staining, and the contents of 5 fungi (including Candida albicans, Aspergillus albicans, Penicillium and Cladosporium) were measured by real-time fluorescence quantitative technique. Cryptococcus sp. Results the results of HE staining showed that the colonic mucosa layer of the model group was almost completely defective, the ulcer formed, the mucosa hyperemia and erosion accompanied by inflammatory cell infiltration, and the treatment group was improved compared with the model group. Compared with the control group, the relative expression levels of Candida albicans, Aspergillus and Penicillium in the other groups were significantly increased, while the relative expression levels of Cladosporium and Cryptococcus were lower than those of the control group (P 0.05). Compared with the model group, the relative expression levels of Candida albicans, Aspergillus and Penicillium in the intestinal tract of rats in each treatment group were significantly decreased, while the relative expression levels of Cladosporium and Cryptococcus were increased (P 0.05). The relative expression of Cryptococcus was superior to that of Anchang decoction in decreasing Aspergillus and increasing Cladosporium, while in high dose group, the relative expression of Candida albicans and Penicillium was decreased in high dose group than in Anchang decoction group. Middle dose group (P 0.05). Conclusion Anchang decoction has a definite regulatory effect on intestinal fungi in rats with ulcerative colitis and may be one of the mechanisms of its therapeutic effect on ulcerative colitis.
