IGFBP3基因内含子2多态性及其与猪主要生长性状的关联分析

【摘要】:胰岛素样生长因子结合蛋白(Insulin-like Growth Factor Binding Proteins,IGFBPs)与动物的生长发育有着密切的联系,为研究IGFBP3的多态性与猪生长性状之间的关系,本实验采用PCR-RFLP方法对大白猪、杜洛克及长白猪IGFBP3基因内含子2的多态性进行分析,并且研究了IGFBP3不同基因型对体长、体高、瘦肉率、活体背膘、肌肉厚度、0~100 kg日增重、30~100 kg日增重及100 kg体重日龄等主要生长性状的影响。结果表明:IGFBP3基因内含子2共417 bp,在3个猪群中有1个A307G多态位点与猪的生长性状有关,并且都是AG基因型占优势,杜洛克与长白猪在该位点上偏离Hardy-Weinberg平衡(P0.010);3个品种猪群IGFBP3基因的多态性信息含量(PIC)均表现为中度多态(0.34~0.37);IGFBP3基因的GG基因型对大白猪的体高(P=0.068)、瘦肉率(P=0.058)的影响较大,对杜洛克猪的瘦肉率(P=0.099)、肌肉厚度(P=0.079)的影响较大,对长白猪的0~100 kg日增重的影响均接近显著水平(P=0.08)。GG基因型猪具有体高、瘦肉率高和生长较快的特点,为以后猪的育种和选种提供参考。
[Abstract]:Insulin-like Growth Factor Binding proteins (IGFBPs) is closely related to the growth and development of animals. In order to study the relationship between IGFBP3 polymorphism and pig growth traits, PCR-RFLP method was used in this study. The polymorphism of intron 2 of IGFBP3 gene in Duroc and Landrace pigs was analyzed, and the effects of different genotypes of IGFBP3 on body length, body height, lean meat rate and backfat in vivo were studied. The effects of muscle thickness of 100 kg / day and 100kg / kg daily gain on the main growth traits. The results showed that the intron 2 of the 1: IGFBP3 gene was 417bp. one A307G polymorphic locus in three breeds was related to the growth traits of pigs, and all of them were the dominant genotype of AG. Duroc and Landrace deviated from the Hardy-Weinberg balance at this locus P0.0101.The polymorphism information content of IGFBP3 gene in the three breeds showed that the GG genotype of IGFBP3 gene had a great influence on the body height of large white pigs, and the lean meat percentage was P0.058. The effect on lean meat rate and muscle thickness of Duroc pigs was greater, and the effects on daily gain of 0 ~ 100kg / kg of Landrace pigs were close to the significant level. The genotype pigs had the characteristics of high body height, high lean meat rate and fast growth. To provide reference for future pig breeding and selection.
