玉米性别决定基因TS2的表达模式分析

【摘要】:[目的]TS2(tasselseed2)是玉米性别决定中的一个关键基因,国内外对其进行了多方面的研究,但其生理学功能至今仍然不清楚。本研究通过TS2基因的表达模式来阐明TS2的表达受什么信号物质刺激,从而推测TS2通过何种生理过程或信号转导途径来实现其在玉米性别决定中的作用。[方法]以玉米自交系B73苗期的叶片、茎尖、根,性别分化时期的穗位叶、雄蕊,开花期的雌蕊、节间、气生根、花丝为试验材料,在V3期(3叶1心期)喷施100μmol·L-1的GA、NAA、ABA、ACC、JA和2.5 mmol·L-1的SA溶液(含0.01%Tween-20),分别于激素处理后0、6、12、24、48、72 h时剪取第3叶中段为样本,采用荧光定量PCR测定TS2受激素诱导的表达量。[结果]TS2基因在已检测的各组织中都有一定量的表达。在叶片中TS2的表达从苗期到开花期逐渐增加,V8期(8叶1心期)叶片内的TS2表达量达到最高峰,以后逐渐下降。苗期(V1~V7)(1叶1心期到7叶1心期)茎尖中TS2的表达量一直很低,但到V8期,植株茎尖开始性别分化时,TS2的表达量迅速提高20多倍。雄穗、雌穗内TS2的表达量高于叶片或其他营养器官。植物激素GA、SA、NAA抑制TS2的表达,ABA、ACC、JA促进TS2的表达,但以JA对TS2的表达影响最大,JA处理后24 h,TS2的表达量提高了84倍。[结论]植物性别决定基因TS2的表达主要受植物生殖发育过程和植物激素茉莉酸信号的调控,预示着TS2的生理功能与植株体内茉莉酸信号转导途径密切相关。
[Abstract]:[objective] TS2tasselseed2 is a key gene in sex determination of maize, which has been studied in many fields at home and abroad, but its physiological function is still unclear. In this study, the expression pattern of TS2 gene was used to elucidate what signal substance stimulates the expression of TS2, so as to speculate the physiological process or signal transduction pathway through which TS2 plays a role in the sex determination of maize. [methods] leaves, stem tips, roots, ear leaves, stamens, pistil, internodes, aerial roots and filaments of maize inbred line B73 at seedling stage were used as experimental materials. 100 渭 mol L-1 GANAA ACCJA and 2.5 mmol L-1 SA solution (containing 0.01 Tween-20) were sprayed on the third leaf of stage V3. The middle segment of the third leaf was cut for 72 h after steroid treatment, and the expression of TS2 induced by hormone was determined by fluorescence quantitative PCR. [results] TS2 gene was expressed in all tissues detected. The expression of TS2 in leaves increased gradually from seedling stage to flowering stage. In seedling stage, the expression of TS2 in stem tip was very low from 1 heart stage to 7 leaf 1 heart stage, but at V8 stage, the expression of Ts2 increased more than 20 times when the stem tip began to differentiate. The expression of TS2 in male ear and female ear was higher than that in leaves or other vegetative organs. The plant hormone GAA SAA inhibited the expression of TS2. ACC-JA promoted the expression of TS2, but JA had the greatest effect on the expression of TS2. 24 h after JA treatment, the expression of TS2 increased 84 times. [conclusion] the expression of plant sex determinant gene TS2 is mainly regulated by plant reproductive development and plant hormone jasmonic acid signal, indicating that the physiological function of TS2 is closely related to the signal transduction pathway of jasmonic acid in plant.
