紫花苜蓿HSC70-2基因克隆及其时空表达

【摘要】:研究应用RNA-seq和聚合酶链式反应确定了一个热休克同源蛋白70-2(MsHSC70-2)基因以及应用荧光定量PCR检测紫花苜蓿不同季节和不同器官中MsHSC70-2的mRNA含量。结果表明:该基因的mRNA序列含有2 025bp,开放阅读框1 950bp,编码649个氨基酸,该基因的蛋白分子量约70.97kDa,等电点4.74。同一器官不同季节MsHSC70-2mRNA含量不同(P0.05),夏季时不同器官间含量存在差异(P0.01),该基因在根、茎、叶和顶芽中的表达趋势从四月份到九月份都是先升高后降低,表明MsHSC70-2的表达受日照长度和温度的调节,其表达量变化影响了各器官的生长发育,高表达促进苜蓿的生长发育,低表达抑制苜蓿的生长发育,该基因表达的降低可能参与秋眠型苜蓿秋眠的发生。研究结果为进一步研究紫花苜蓿HSC70-2的功能提供了依据。
[Abstract]:A heat shock homologous protein 70-2MsHSC70-2) gene was determined by RNA-seq and polymerase chain reaction. The mRNA content of MsHSC70-2 in different seasons and organs of alfalfa was detected by fluorescence quantitative PCR. The results showed that the mRNA sequence of the gene contained 2 025 BP, open reading frame 190 BP, encoding 649 amino acids, the molecular weight of the protein was about 70.97 kDa, and the isoelectric point was 4.74. The content of MsHSC70-2mRNA was different in different seasons of the same organ, and the content of P0.01 was different among different organs in summer. The expression trend of this gene in root, stem, leaf and terminal bud increased first and then decreased from April to September. The results showed that the expression of MsHSC70-2 was regulated by sunshine length and temperature. The changes of MsHSC70-2 expression affected the growth and development of various organs, the high expression promoted the growth and development of alfalfa, and the low expression inhibited the growth and development of alfalfa. The decrease of the gene expression may be involved in the autumn dormancy of alfalfa. The results provided a basis for further study on the function of alfalfa HSC70-2.
