利用广谱抗病基因Pigm改良水稻稻瘟病抗性

【摘要】:为改良已选育的高产优质粳稻高代新品系的稻瘟病抗性,以引进的携带有水稻广谱抗稻瘟病基因Pigm的籼稻品种谷梅4号和粳稻材料MP3为供体,利用花粉管通道技术将其基因组DNA导入到SD-2、SD-3、SD-4、SD-5、SD-6、DJ13、ZD15、14PB1和14PB4等高代品系和品种宁粳43号中,利用Pigm基因的显性标记M26205和M80410对受体及获得的导入系进行检测,结果表明:在所有受体中,两个标记均未扩增出条带,说明所有受体本身都不含Pigm基因;在检测的854株导入株系中,利用标记M26205检测总共有328株能够扩增出856 bp,或1 019 bp和856 bp的条带,初步推断这328个株系携带有Pigm基因,平均阳性株率为40.51%;利用标记M80410检测总共有169株能够扩增出517 bp的条带,推断这169个株系可能携带有Pigm基因,平均阳性株率为19.83%。选择两个标记能同时检测出的阳性植株107株作为Pigm基因的导入系,进行进一步的加代培育和辅助选择。
[Abstract]:In order to improve the blast resistance of the new high yield and good quality japonica rice lines, the introduced indica rice cultivar Gumei 4 carrying the broad-spectrum blast resistance gene Pigm and the japonica rice material MP3 were used as donors. The genomic DNA was introduced into high generation lines and cultivar Ningjing 43 by pollen tube pathway technique. The results showed that: among all the receptors, the dominant markers M26205 and M80410 of the Pigm gene were used to detect the receptors and the resulting lines. No bands were amplified from the two markers, indicating that none of the receptors contained Pigm gene, and a total of 328 of the 854 lines were detected by using marker M26205 to amplify 856 BP, or 1 019 BP and 856 BP bands. It was preliminarily inferred that these 328 strains carried Pigm gene, and the average positive rate was 40.51.The results showed that 169 strains could amplify 517bp bands by using marker M80410. It was inferred that the 169 lines might carry Pigm gene, and the average positive rate was 19.833%. 107 positive plants which could be detected simultaneously by two markers were selected as Pigm gene importing lines for further generation and selection.
