宁夏地区回汉儿童HLA基因多态性研究及其与体质和紫癜的相关性

【摘要】:目的 探索宁夏地区回汉儿童的HLA-B、DRB1等位基因多态性,通过计算、分析宁夏地区回汉儿童HLA-B、DRB1等位基因的分布特点,并与中医体质进行统计分析,以探讨HLA基因多态性与中医体质之间的关联,同期与其他人群的相关等位基因做比较,期待为本地区儿童过敏性紫癜及紫癜肾的发病及转归预测提供客观的实验室依据。方法 采集宁夏地区76名回汉儿童的血样,通过PCR-序列特异性引物(PCR-SSP)方法,对HLA-B、DRB1基因分型进行检测,并对分型结果进行统计分析,同期与其它人群数据做比较。与此同时,我们对研究对象进行中医体质的划分,并且将HLA数据与中医体质进行统计分析。结果 1.宁夏汉族HLA-B位点中共检测到等位基因19种,基因频率由高到低分别为B*15(16.3%)、B*35、40(13.0%)、B*13、46(9.8%)、B*44、51(5.4%)、B*50(4.3%)、B*07、37、48、52(3.3%)、B*08、55(2.2%)、HLA-B*18、38、49、54、57(1.1%)。HLA-DRB1位点中共检测到等位基因13种。基因频率由高到低分别为DRB1*12(19.6%)、DRB1*09(16.3%)、DRB1*07(12.0%)、DRB1*04、14(9.8%)、DRB1*15(8.7%)、DRB1*08(7.6%)、DRB1*13(4.3%)、DRB1*11、16(3.3%)、DRB1*03、10(2.2%)、DRB1*01(1.1%)。2.宁夏回族HLA-B位点中共检测到等位基因19种,基因频率由高到低分别为B*40、46(18.3%)、B*15(11.7%)、B*13(8.3%)、B*07、35、44、48、51(5.0%)、B*58(3.3%)、B*08、37、38、39、45、49、52、54、55(1.7%)。HLA-DRB1位点中共检测到等位基因12种,基因频率由高到低分别为DRB1*12(16.7%)、DRB1*15(15.0%)、DRB1*04、14(13.3%)、DRB1*11(11.7%)、DRB1*07、09、13(6.7%)、DRB1*03、08(3.3%)、DRB1*01、16(1.7%)。3.在宁夏汉族、回族人群中,HLA-B、HLA-DRB1等位基因频率分布基本相似,差异无统计学意义。4.宁夏汉族人群的HLA-B、DRB1等位基因与山西汉族、山东汉族人群的分布情况基本相似,但是又具有自身的分布特点。结论 1.宁夏汉族与回族人群HLA-B、HLA-DRB1等位基因分布基本相似,差异无统计学意义;2.宁夏汉族人群HLA-B、HLA-DRB1等位基因与山西汉族、山东汉族基因频率的分布大致相同,但又有其自身的分布特点;3.可以推测HLA-DRB1*01/11为宁夏汉族HSP/HSPN患儿的易感基因,HLA-DRB1*07为抵抗基因;HLA-B*35、HLA-DRB1*01为宁夏回族HSP/HSPN患儿的易感基因,HLA-DRB1*07为抵抗基因;4.HLA具有明显的种族和地域性差异,为群体遗传学和HLA疾病相关性研究提供了重要的参考资料。
[Abstract]:Objective to explore the polymorphism of HLA-BmDRB1 alleles in Hui Han children in Ningxia, and to analyze the distribution characteristics of HLA-BnDRB1 alleles in Hui Han children in Ningxia, and to make statistical analysis with TCM constitution. In order to explore the association between HLA gene polymorphism and TCM physique, and to compare with other alleles in the same period, we hope to provide an objective laboratory basis for predicting the pathogenesis and prognosis of Henoch-Schonlein purpura and Henoch-Schonlein purpura kidney in children in this area. Methods Blood samples were collected from 76 Hui Han children in Ningxia area. The genotyping of HLA-BmDRB1 gene was detected by PCR-SSPP with PCR- sequence specific primers, and the typing results were statistically analyzed, and compared with other population data at the same time. At the same time, we classify the subjects of TCM constitution, and make statistical analysis of HLA data and TCM constitution. Result 1. A total of 19 alleles were detected at HLA-B locus in Han nationality in Ningxia. The frequencies of the alleles from high to low were BH151616.33B, B3554040 and 13.013.00.There were 13 alleles detected at B0737777777777772M3.3B and B08552.2in B08552.2and B08552.2B08552.2at B08552.2B, respectively. The gene frequencies from high to low were DRB1121219.6C and DRB10916.330.There was DRB10712.00.DRB1014149.8.
