水稻OsRhoGAP1基因的序列特征及表达特性分析

【摘要】:Os Rho GAP1是本实验室前期从水稻幼穗c DNA文库中分离获得的功能未知基因。通过生物信息学和现代分子生物学技术,系统探讨了水稻Os Rho GAP1的组织表达特性和不同激素处理下的表达情况。生物信息学分析表明,Os Rho GAP1具有植物Rho GAP蛋白典型的结构特征,即CRIB基序和Rho GAP结构域。q RT-PCR检测结果显示该基因的表达具有时空特异性,幼苗期主要在根和叶中表达,幼穗期主要在幼穗和叶中表达,成熟期则主要在根中表达。同时,经ABA、SA、GA和Me JA处理后,Os Rho GAP1基因表达量有明显上调,经IAA和6-BA处理后,Os Rho GAP1基因表达量出现显著下调,表明该基因的表达受到多种激素调控。
[Abstract]:Os Rho GAP1 is an unknown gene isolated from rice panicle cDNA library in early stage. The tissue expression characteristics of rice Os Rho GAP1 and the expression of Os Rho GAP1 under different hormone treatments were systematically studied by bioinformatics and modern molecular biology techniques. Bioinformatics analysis showed that OOS Rho GAP1 had the typical structural characteristics of plant Rho gap protein. The results of CRIB motif and Rho gap domain. Q RT-PCR showed that the gene expression was space-time specific, and was mainly expressed in roots and leaves at seedling stage. The expression was mainly in the young ears and leaves at the spike stage and in the root at the mature stage. At the same time, the expression of OS-Rho GAP1 gene was up-regulated by GA and me JA, and down-regulated by IAA and 6-BA, indicating that the expression of GAP1 gene was regulated by many hormones.
