柑橘冷诱导基因低温响应及转化柠檬

【摘要】:本课题组已经从耐寒性柑橘枳中克隆获得一个与低温锻炼有关的基因,并将其命名为Ptcor8。同时发现该基因在金柑、柠檬等柑橘种类中具有高度同源性,而且该同源基因表达与柑橘抗寒能力大小呈现相同趋势,枳最耐寒,能耐受-26℃低温,柠檬最不耐寒,枳中的表达量是柠檬的25倍。另外研究比对不同柑橘种类中同源基因启动子的序列发现,枳启动子与柠檬启动子的同源性为88.17%,并且枳启动子含有3个与逆境响应相关的特有调控元件。于是构建了含有启动子和cor8基因的融合植物表达载体,用农杆菌介导的瞬时表达法,将含有不同载体的菌液注射到墨西哥莱檬叶片中,进行瞬时表达,发现枳启动子对低温响应的敏感度更高。在此条件下,将pPtcor8::Ptc-or8::YFP, pPtcor8::Clcor8::YFP, pClcor8::Ptcor8::YFP3个植物表达载体转入到不耐寒的柠檬,并对柠檬遗传转化条件进行优化,筛选出最佳组合的再生条件和筛选压,最终得到了转基因植株本研究取得了以下主要结果:1、将构建好的4个植物表达载体(35S::Ptcor8::YFP, pPtcor8::Ptcor8::YFP, pClcor8::Ptcor8::YFP,pPtcor8::Clcor8::YFP)采用农杆菌介导的瞬时表达法注射墨西哥莱檬叶片,YFP报告基因的荧光检测说明枳的启动子比柠檬启动子在低温下更敏感。2、对柠檬遗传转化条件进行优化,在再生培养基中不同浓度的6-BA,卡那霉素以及除草剂进行筛选,得出了再生培养基为MT+0.75 mg/L 6-BA,添加75 mg/LKan或者10mg/LBastao3、将pPtcor8::Ptcor8::YFP, pPtcor8::Clcor8::YFP, pClcor8::Ptcor8::YFP3个植物表达载体转入到不耐寒的柠檬,得到含有ppPtcor8::Ptcor8::YFP抗性芽34个,pClcor8::Ptcor8::YFP抗性芽48个,pPtcor8::Clcor8::YFP抗性芽29个,通过报告基因的荧光检测得到成功转入pPtcor8::Ptcor8::YFP, pPtcor8::Ptcor8::YFP, pClcor8:: Ptcor8::YFP,pPtcor8::Clcor8::YFP的柠檬植株分别是3株,4株,2株;并移栽于温室大棚。
