黄芪多糖对肌源性干细胞Wnt信号相关基因表达的影响

【摘要】:目的 探讨黄芪多糖干预的肌源性干细胞Wnt/β-catenin信号通路的相关基因表达的变化。方法 将经混合酶消化法、多次差速贴壁法分离纯化的MDSCs随机分为6组,进行实验干预,即空白对照组、APS组、Wnt激动剂组、APS+Wnt激动剂组、Wnt抑制剂组及APS+Wnt抑制剂组。Realtime PCR检测MDSCs Wnt信号通路上下游节点基因Wnt3、β-catenin及MDSCs增殖分化相关基因CyclinD1、c-myc、CD34mRNA的表达变化。结果 流式细胞仪检测结果表明MDSCs CD34、Sca-1阳性。倒置显微镜下观察,APS组、Wnt激动剂组、APS+Wnt激动剂组MDSCs较空白对照组生长密集,APS+Wnt激动剂组聚团明显,Wnt抑制剂组MDSCs增殖明显受到抑制。PCR检测结果显示:APS+激动剂干预组的Wnt信号通路各节点基因表达水平最高,Wnt激动剂组与APS组其次,空白对照组最低。结论 在黄芪多糖的干预作用下,持续激活Wnt信号通路能够促进MDSCs的增殖分化。
[Abstract]:Objective to investigate the expression of Wnt / 尾-catenin signaling pathway in myogenic stem cells treated with astragalus polysaccharide. Methods MDSCs isolated and purified by mixed enzyme digestion and multiple differential adherence were randomly divided into 6 groups for experimental intervention. Wnt agonist group and APS Wnt inhibitor group. Realtime PCR was used to detect the expression of Wnt gene Wnt3, 尾 -catenin and CyclinD1c-mycCD34 mRNA in MDSCs Wnt signaling pathway. Results the results of flow cytometry showed that MDSCs CD34 and Sca-1 were positive. Observation of MDSCs in APS Wnt agonist group under inverted microscope compared with APS Wnt agonist group in blank control group the proliferation of MDSCs in APS Wnt agonist group was significantly inhibited. PCR results showed that the proliferation of MDSCs was significantly inhibited in the control group treated with APS Wnt agonist. PCR results showed that the MDSCs were significantly inhibited in the control group treated with the APS agonist. The highest level of gene expression in each node of Wnt signaling pathway was found in Wnt agonist group and APS group. Blank control group was the lowest. Conclusion the continuous activation of Wnt signaling pathway can promote the proliferation and differentiation of MDSCs under the intervention of astragalus polysaccharide.
