偏头痛模型大鼠降钙素基因相关肽、腺苷A1、A2a受体的表达及天舒胶囊对其影响

【摘要】:目的研究降钙素基因相关肽(calcitonin gene-related peptide,CGRP)、腺苷 A1、A2a受体(A1R、A2aR)在偏头痛模型大鼠三叉神经节(trigeminal ganglion,TG)和三叉神经脊束尾核(spinal trigeminal nucleus caudalis, TNC)中的表达及天舒胶囊对其表达的影响。探讨这三种物质在偏头痛大鼠模型中的表达特点和相互关系及天舒胶囊预防偏头痛的可能作用机制。方法将40只大鼠随机分为空白对照组(BC组,n=10)、假手术组(SO组,n=10)、电刺激三叉神经节组(electrical stimulation of the trigeminal ganglion,ESTG 组,n=10)、天舒胶囊干预组(TC组,n=10)。空白组处死前不做特殊处理,ESTG组电刺激三叉神经节30min, SO组只插电针不给予电刺激,刺激时间同ESTG组,TC组大鼠进行ESTG造模前按体质量3.75 g/(kg·d)给予天舒胶囊灌胃,每天1次,持续7 d。最后1次灌胃后lh建立ESTG模型,模型复制方法同ESTG组。模型成功后,立即断头处死大鼠,快速取出大鼠的刺激侧的TG及TNC所在的脑干部位,-80℃冰箱保存备用。采用REAL-TIME PCR、Western Blot技术分别检测4组大鼠TG、TNC中CGRP、A1R、A2aR的表达量的变化。结果①在TG和TNC中,偏头痛模型组大鼠的CGRPmRNA、A2aRmRNA的表达量高于空白对照组和假手术组(P0.05);偏头痛模型组大鼠A1RmRNA表达量低于空白对照组和假手术组(P0.05)空白组和假手术组之间此三种物质的表达量差异没有统计学意义(P0.05)。②在TG和TNC中,偏头痛模型组大鼠的CGRP、A2aR的蛋白表达量高于空白组和天舒胶囊干预组大鼠(P0.05), A1R的蛋白表达量低于空白组和天舒胶囊干预组(P0.05);空白组和天舒胶囊干预组之间此三种物质的表达量差异没有统计学意义(P0.05)。③CGRP、A2aR、A1R三种物质之间的表达量的关系。在偏头痛大鼠TG和TNC中,CGRP和A2aR两者之间均存在正相关关系(P0.01);而CGRP和A1R两者之间存在负相关关系(P0.01)。结论①CGRP、A2aR在偏头痛大鼠模型中高表达,A1R低表达,参与偏头痛的病理生理进程。②预防应用天舒胶囊影响CGRP、A2aR、A1R的表达,对偏头痛起到一定保护作用。③CGRP同A2aR、A1R之间存在相关关系,三种物质之间相互作用参与偏头痛的发病过程。
[Abstract]:Objective to study the expression of calcitonin gene-related peptide (calcitonin gene-related peptidea) and adenosine A1RtA2a receptor (A1R- A2aR) in trigeminal ganglion TG and (spinal trigeminal nucleus caudalis, TNC) of caudate nucleus of spinal tract of trigeminal nerve in migraine model rats and the effect of Tianshu capsule on the expression of CGRP. To investigate the expression and relationship of these three substances in migraine rat model and the possible mechanism of Tianshu capsule in preventing migraine. Methods Forty rats were randomly divided into three groups: blank control group (BC group), sham operation group (so group), (electrical stimulation of the trigeminal ganglion stimulation group (ESTG group) and Tianshu capsule intervention group (TC group). In the blank group, the trigeminal ganglion was stimulated 30 minutes without special treatment before death, the so group was treated with electroacupuncture only, and the stimulation time was the same as that in the ESTG group before the ESTG model was made. Tianshu capsule was given orally according to the body mass of 3.75 g / (kg d), once a day for 7 days. The ESTG model was established at lh after the last gastric perfusion, and the model was duplicated with the same method as that in the ESTG group. After the model was successfully established, the rats were killed immediately, and the stimulation side TG and the brain stem of TNC were quickly removed and stored in refrigerator at -80 鈩,
