TRV介导的VIGS技术在果蔬基因功能研究中的应用

【摘要】:病毒诱导的基因沉默(virus-induced gene silencing,VIGS)是近年来开发的一种快速鉴定植物基因功能的反向遗传学技术。因该技术需要借助病毒载体来实现,所以病毒载体的开发一直备受关注。在已开发的病毒载体中,烟草脆裂病毒(Tobacco rattle virus,TRV)具有寄主范围广、沉默效率高、可在多个组织产生沉默,并且病毒诱导的症状轻微等独特的优势。因此,TRV-VIGS技术已成功用于多种果蔬作物基因功能的研究。本文就TRV载体的结构特点、作用机制、在果蔬基因功能研究中的应用及存在的问题等进行了分析和总结。
[Abstract]:Virus-induced gene silencing (virus-induced gene silencing) is a rapid reverse genetic technique for the identification of plant gene function in recent years. Because the technology needs to be realized by virus vector, the development of virus vector has been paid more and more attention. Among the virus vectors developed, tobacco embrittlement virus (Tobacco rattle virustrv) has many unique advantages, such as wide host range, high silencing efficiency, silencing in multiple tissues and mild symptoms induced by the virus. Therefore, TRV-VIGS technology has been successfully used to study the gene function of many fruits and vegetables. In this paper, the structure and mechanism of TRV vector, its application in the study of gene function of fruits and vegetables and the existing problems are analyzed and summarized.
