干扰素刺激基因在鸡传染性支气管炎病毒感染雏鸡气管黏膜中表达的动态变化

【摘要】:为了解鸡干扰素刺激基因(ISG)在鸡传染性支气管炎病毒(IBV)感染雏鸡气管黏膜中的表达情况,采用实时荧光定量PCR检测IBV M41株感染SPF鸡后不同时相气管黏膜中β干扰素(IFN-β)和干扰素刺激基因mRNA转录水平和IBV病毒载量变化。结果显示,鸡气管黏膜中IFN-β和干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5mRNA转录水平在感染后(DPI)第1~14天显著(P0.05)或极显著(P0.01)上调,且均在5DPI达到峰值;鸡气管黏膜中IBV病毒载量在1DPI急剧升高,在5DPI达到峰值,14DPI病毒载量急剧下降。结果表明,IFN-β、干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5mRNA转录水平与IBV病毒载量变化趋势一致,提示IFN-β、干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5在抵御IBV入侵、抑制IBV复制、增殖以及病毒清除中发挥重要作用。试验结果丰富了宿主抗IBV感染的固有免疫应答机制,为IBV感染的预防和控制提供了理论依据。
[Abstract]:To investigate the expression of chicken interferon stimulating gene (ISG) in tracheal mucosa of chickens infected with avian infectious bronchitis virus (IBV). The levels of interferon 尾 (IFN- 尾), IFN- stimulated gene mRNA transcription and IBV viral load in tracheal mucosa of IBV M41 strain infected with SPF chicken were detected by real-time fluorescence quantitative PCR. The results showed that the transcription levels of IFN- 尾 and interferon stimulating genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5mRNA in tracheal mucosa of chicken were significantly up-regulated (P0.05) or extremely significant (P0.01) on the 1st day after (DPI) infection, and both reached the peak in 5DPI, the viral load of IBV in chicken tracheal mucosa increased sharply in 1DPI. At the peak of 5DPI, the viral load of 14DPI decreased sharply. The results showed that the transcriptional levels of IFN- 尾, IFN- stimulated genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5mRNA were consistent with the changes of IBV viral load, suggesting that IFN- 尾, IFN- stimulated genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5 play an important role in resisting IBV invasion, inhibiting IBV replication, proliferation and virus clearance. The results enrich the innate immune response mechanism of host against IBV infection and provide a theoretical basis for the prevention and control of IBV infection.
