基因芯片技术研究黄秦艽对雪上一枝蒿急性心脏毒性的作用机制

【摘要】:目的:利用芯片技术探讨黄秦艽对雪上一枝蒿所致小鼠心脏急性毒性基因表达的影响及分子机制.方法:25只雄性昆明小鼠随机分为正常对照组、雪上一枝蒿氯仿部位组(CFA)和黄秦艽水提物组(WVBF),通过表达谱对各组心脏差异表达基因进行生物信息学研究.结果:WVBF对CFA诱导的急性心脏毒性具有明显的解毒效果,染毒组与正常组相比,差异基因的功能主要涉及机体内物质的异常代谢,WVBF主要通过干扰CFA引起的内质网应激反应及线粒体功能紊乱过程,提高细胞存活率,使心肌恢复正常功能.结论:黄秦艽可以通过多条信号通路缓解雪上一枝蒿所致的小鼠心肌毒性作用.
[Abstract]:Aim: to investigate the effect of Gentiana macrophylla Gentiana on the expression of acute toxic genes in heart of mice induced by Artemisia mongolicum by microarray technique and its molecular mechanism. Methods: Twenty-five male Kunming mice were randomly divided into normal control group, (CFA) group and Gentiana macrophylla water extract group (WVBF),). The differentially expressed genes in each group were studied by bioinformatics. Results: WVBF had obvious detoxification effect on acute cardiotoxicity induced by CFA. The function of differentially expressed genes is mainly related to the abnormal metabolism of substances in the body. WVBF improves the survival rate of cells by interfering with the stress response of endoplasmic reticulum and mitochondrial dysfunction induced by CFA, and restores the normal function of myocardium. Conclusion: Gentiana macrophylla can attenuate the myocardial toxicity induced by Artemisia mongolica through several signal pathways.
