Panx1基因过表达对人肝癌Hep3B细胞增殖及迁移的影响

【摘要】:目的探讨Panx1基因过表达对人肝癌Hep3B细胞增殖及迁移的影响。方法构建Panx1过表达慢病毒质粒和过表达空载慢病毒质粒,包装生产慢病毒,分别转染Hep3B细胞(过表达组、对照组)。用Western blotting法检测Hep3B细胞Panx1、上皮钙黏蛋白(E-cadherin)及波形蛋白(Vimentin)表达,CCK-8法检测细胞增殖能力(OD值),划痕实验检测细胞迁移能力(相对迁移距离、划痕愈合率)。结果过表达组Panx1、E-cadherin蛋白相对表达量较对照组高(P均0.05),Vimentin蛋白相对表达量较对照组低(P0.05);各时间点(0 h除外)OD值较对照组低(P均0.05);相对迁移距离较对照组低,划痕愈合率较对照组高(P均0.05)。结论 Panx1基因过表达可抑制Hep3B细胞增殖与迁移。
[Abstract]:Objective to investigate the effect of overexpression of Panx1 gene on proliferation and migration of human hepatoma Hep3B cells. Methods Panx1 overexpression lentivirus plasmids and empty lentivirus plasmids were constructed and packaged to produce lentivirus. The lentivirus was transfected into Hep3B cells (overexpression group, control group). The expression of Panx1, epithelial cadherin (E-cadherin) and vimentin (Vimentin) was detected by Western blotting assay, cell proliferation ability (OD value) was detected by CCK-8 assay, and migration ability (relative migration distance, scratch healing rate) was detected by scratch test. Results the relative expression of Panx1,E-cadherin protein in the overexpression group was higher than that in the control group (P < 0. 05), the OD value at each time point (except 0 h) was lower than that in the control group (P 0. 05), and the relative expression of), Vimentin protein in the overexpression group was lower than that in the control group (P 0. 05). The relative migration distance was lower than that of control group, and the rate of scratch healing was higher than that of control group (P 0.05). Conclusion overexpression of Panx1 gene can inhibit the proliferation and migration of Hep3B cells.
