牦牛IL-1β基因的鉴定及组织表达

【摘要】:为了克隆麦洼牦牛IL-1β基因并分析其蛋白质结构及表达,给研究IL-1β基因的生物学功能提供理论基础,试验利用RT-PCR方法扩增麦洼牦牛IL-1β基因序列,分析IL-1β基因在不同组织中的表达情况,通过生物学软件进行核酸序列分析及蛋白质结构预测。结果表明:麦洼牦牛IL-1β基因大小为899 bp,Gen Bank登录号为KU510225。此序列编码266个氨基酸,蛋白质分子质量为30 684.0 u,不含跨膜区且没有信号肽;蛋白二级结构以α-螺旋为主,三级结构与小鼠IL-1β蛋白结构类似。IL-1β基因在各组织中表达量的顺序为脾脏肝脏心脏肺脏肾脏。说明试验成功克隆了麦洼牦牛IL-1β基因,在脾脏表达量高,表明IL-1β基因可能参与机体的免疫应答。
[Abstract]:In order to clone and analyze the protein structure and expression of IL-1 尾 gene in Maiwa yak, and to provide a theoretical basis for studying the biological function of IL-1 尾 gene, RT-PCR was used to amplify the IL-1 尾 gene sequence of Mai WA yak. The expression of IL-1 尾 gene in different tissues was analyzed. Nucleic acid sequence analysis and protein structure prediction were carried out by biological software. The results showed that the size of IL-1 尾 gene of Maiwa yak was 899 bp,Gen Bank and the accession number was KU510225.. This sequence encodes 266 amino acids with a molecular weight of 30 684.0 u, no transmembrane region and no signal peptide. The secondary structure of the protein was 伪 -helix, and the tertiary structure was similar to that of mouse IL-1 尾 protein. The order of expression of IL-1 尾 gene in each tissue was spleen, liver, heart, lung and kidney. The results showed that the IL-1 尾 gene of Maiwa Yak was cloned successfully and expressed in the spleen, indicating that the IL-1 尾 gene might be involved in the immune response of the body.
