姜黄素对结肠癌细胞株NKD2及CXCR4基因表达影响的研究

【摘要】:[目的]观察姜黄素对SW620细胞及裸鼠NKD2、CXCR4表达水平的影响。[方法]通过MTS检测姜黄素对SW620细胞活性的影响;运用Western blot、Real-time q PCR检测不同浓度姜黄素对SW620细胞及转染NKD2 si RNA后SW620细胞内NKD2、CXCR4蛋白和m RNA表达的影响。雄性BALB/c裸鼠8只,随机分成对照组与姜黄素组,每组4只。采用皮下接种SW620细胞。造模后第7d开始给对照组与姜黄素组分别注射对照液和100mg/kg·d浓度的姜黄素悬液,连续用药14d。给药后检测瘤体体积,通过Real-time q PCR检测瘤体组织内NKD2及CXCR4表达水平。[结果 ]姜黄素能明显抑制SW620细胞的增殖活性,显著性上调SW620细胞内NKD2表达及下调CXCR4的表达,且呈剂量依赖性。转染后发现能上调姜黄素处理后的肿瘤细胞CXCR4的表达水平。体内实验表明姜黄素抑瘤率为30.69%。通过q PCR发现瘤体组织中NKD2的表达量增加,CXCR4的表达量显著性降低。[结论]姜黄素均能从体内外明显抑制结肠癌细胞SW620的生长,其作用机制可能通过上调抑制基因NKD2的表达,抑制Wnt信号通路,进而下调肿瘤细胞CXCR4表达,从而起到抑制肿瘤侵袭转移的作用。
[Abstract]:[objective] to observe the effect of curcumin on the expression of NKD2,CXCR4 in SW620 cells and nude mice. [methods] the effect of curcumin on the activity of SW620 cells was detected by MTS, and the expression of NKD2,CXCR4 protein and m RNA in SW620 cells and SW620 cells after NKD2 si RNA transfection was detected by Western blot,Real-time Q PCR. Eight male BALB/c nude mice were randomly divided into control group and curcumin group with 4 rats in each group. SW620 cells were inoculated subcutaneously. The control group and curcumin group were injected curcumin suspension with control solution and 100mg/kg d concentration respectively on the 7th day after modeling. The volume of tumor was measured after administration, and the expression of NKD2 and CXCR4 in tumor tissue was detected by Real-time Q PCR. [results] curcumin significantly inhibited the proliferation of SW620 cells, significantly up-regulated the expression of NKD2 and down-regulated the expression of CXCR4 in SW620 cells in a dose-dependent manner. After transfection, it was found that curcumin could up-regulate the expression of CXCR4 in tumor cells treated with curcumin. The inhibition rate of curcumin was 30.69% in vivo. Q PCR showed that the expression of NKD2 increased and the expression of CXCR4 decreased significantly. [conclusion] curcumin can obviously inhibit the growth of SW620 in colon cancer cells in vitro and in vivo. The mechanism of curcumin can inhibit the expression of Wnt signal pathway by up-regulating the expression of NKD2 gene, and then down-regulate the expression of CXCR4 in tumor cells. Thus it can inhibit the invasion and metastasis of tumor.
