唐菖蒲GhMYC2基因启动子的活性分析

【摘要】:以唐菖蒲(Gladiolus hybridus)品种"Rose Supreme"为材料,采用hiTAIL-PCR方法克隆得到GhMYC2基因上游906bp的启动子序列。利用植物顺式作用元件查询数据库PLACE分析发现,GhMYC2启动子序列中不仅含有响应茉莉酸(jasmonate,JA)的元件T/GBOXATPIN2,还含有大量与病原菌诱发因子表达调节相关的转录因子WRKY以及抗性元件W-box、E-box等顺式作用元件。在ProGhMYC2::GUS转基因拟南芥中发现,GhMYC2的表达具有时空特异性,且主要在根、叶中表达。对转基因拟南芥进行茉莉酸甲酯(methyl jasmonic acid,MeJA)和灰霉菌(Botrytis cinerea)处理发现,转基因植株中GUS的表达均上调,表明其表达受到MeJA和灰霉菌的影响,此结果可为研究JAs途径调控植物抵御病害提供一定的参考。
[Abstract]:Using Gladiolus (Gladiolus hybridus) variety "Rose Supreme" as the material, the promoter sequence of GhMYC2 gene upstream 906bp was cloned by hiTAIL-PCR method. The PLACE analysis of plant cis-acting element query database showed that the GhMYC2 promoter sequence not only contains the element T / GBOX ATP IN2, which is responsive to jasmonate,JA, but also contains the component T / GBOX ATP-IN2, which is responsible for jasmonic acid (jasmonate,JA). There are also a large number of transcription factor WRKY related to the expression regulation of pathogen inducing factors and cis-acting elements such as the resistance element W-box E-box and so on. In ProGhMYC2::GUS transgenic Arabidopsis thaliana, it was found that the expression of GhMYC2 was space-time specific and mainly expressed in roots and leaves. The expression of GUS was up-regulated in transgenic Arabidopsis thaliana treated with methyl jasmonate (methyl jasmonic acid,MeJA) and (Botrytis cinerea), indicating that the expression of GUS was affected by MeJA and Ash. These results may provide some references for the study of plant disease resistance by JAs pathway.
