拟南芥开花抑制基因TFL1的蛋白表达及纯化

【摘要】:TFL1基因是植物中的一个开花抑制基因,与促进植物开花的FT是同源基因,但是二者发挥相反的功能。本研究为了获得TFL1融合蛋白,首先将TFL1基因构建到PGEX-4T-3表达载体上,获得TFL1基因的原核表达载体PGEX-4T-3-GST-TFL1,转化大肠杆菌表达菌株BL-21(DE3)。在37℃,0.5mmol/L IPTG条件下诱导出了大小为46kD的GST-TFL1融合蛋白。经过GST纯化基质纯化后,获得了质量较高、并且能被TFL1抗体特异识别的GST-TFL1蛋白,为进一步研究TFL1的互作蛋白及其抑制植物开花的分子机制提供了一个十分有力的工具。
[Abstract]:TFL1 gene is a flowering suppressor gene in plants. It is a homologous gene with FT which promotes flowering in plants, but they play an opposite function.
