4个线粒体基因在鲤所属国家种质鉴定中的应用及效果评价

【摘要】:近年来,mtDNA条形码技术已广泛应用于鱼类种质鉴定研究。为探讨该技术在鲤种质鉴定方面应用的可行性及如何有效应用于地域种群等鉴定分析,从NCBI网站(http://www.ncbi.nlm.nih.gov/)搜集鲤COⅠ、COⅡ、Cytb和D-loop4个mtDNA序列,并尝试基于DNA条形码技术来进行鲤所属国家的鉴定分析。结果表明:Cytb相对其他3个线粒体序列存在较大的DNA条形码间隙,COⅡ基因最佳匹配的准确率最高(准确率82.97%),基于4个mtDNA序列的聚类分析表明鲤所属国家按照临近区域聚类。综合条形码间隙及假阳性率和假阴性率的评价,研究认为,相比其他3个mtDNA序列,Cytb基因用于鉴定鲤所属国家更具优势,但其鉴定效率仍不理想。综上所述,mtDNA序列用于鉴定鲤不同地域的技术依然需要改进,以降低这一技术的假阳性率和假阴性率。
