黄连素调节BMP4转录通路基因表达改善2型糖尿病地鼠内脏白色脂肪组织胰岛素抵抗的研究

【摘要】:研究黄连素对2型糖尿病地鼠内脏白色脂肪组织(VWAT)中BMP4转录通路和白/棕脂组织转换通路基因mRNA表达的影响并探讨相关机制。应用高脂饮食诱导肥胖胰岛素抵抗地鼠模型,给予小剂量链脲菌素建立2型糖尿病地鼠模型。造模结束后随机分成正常对照组、肥胖胰岛素抵抗组、2型糖尿病组和2型糖尿病黄连素治疗组。治疗9周后,应用实时定量PCR方法检测各组地鼠VWAT中BMP4通路、白/棕脂组织转换通路及靶基因的mRNA表达改变。结果显示,模型地鼠VWAT中BMP4,BMPRⅡ,BMPRl A,Smad1,Smad5,Smad8,p38/MAPK,ATF2,PRDM16,C/EBPβ,PGC1α,PPARγ及棕脂组织特异基因的mRNA表达降低,而Smad6,Smurf1和白脂组织特异基因的mRNA表达增加。黄连素治疗调节VWAT中BMP4转录通路和棕脂组织转录通路,诱导棕脂组织特异基因mRNA的表达,诱导白色脂肪棕色化基因表型,改善脂诱性胰岛素抵抗。结果表明,BMP4转录通路参与脂诱性内脏白色脂肪组织胰岛素抵抗(FIVWATIR)的形成及黄连素诱导白色脂肪棕色化的分子机制。

The obese insulin resistant hamster model was induced by high fat diet and the type 2 diabetic hamster model was established by low dose streptozotocin.
