亚洲玉米螟滞育期间耐寒相关基因的表达分析

【摘要】:亚洲玉米螟Ostrrinia furnacalis(Guenee),是我国主要玉米害虫,属典型的兼性滞育昆虫,通常以5龄低温耐受型幼虫滞育越冬。为探索亚洲玉米螟滞育幼虫受到低温胁迫时,耐寒相关基因的表达量变化规律,本文在评估筛选亚洲玉米螟越冬幼虫及不同虫态内参基因表达稳定性的基础上,选取编码耐寒相关蛋白的基因—精氨酸激酶基因(AK),谷胱甘肽转移酶基因(Gst)和磷酸甘油醛异构酶基因(Tpi),研究热驯化(20℃)与冷驯化(5℃)维持滞育状态下及在越冬期间的滞育幼虫的AK、Gst和Tpi基因表达的时间动态,旨在为解析亚洲玉米螟幼虫对低温胁迫的分子适应机制奠定基础。全文试验结果如下:(1)筛选出可用于研究亚洲玉米螟滞育时基因表达研究的最适内参基因。亚洲玉米螟不同虫态下,4个候选内参基因的稳定性大小排序为18S rRNAEF1-αβ-ctinPS3;越冬滞育幼虫中,稳定性大小排序为β-aactinEF1-α18S rRNARPS3。在亚洲玉米螟不同虫态和越冬滞育幼虫中,18SrRNA和β-actin可分别作为目的基因表达水平分析试验中的内参基因。(2)探索亚洲玉米螟滞育相关的胁迫耐受性过程。分析AK、Gst和Tpi基因在确定滞育、热驯化(20℃)与冷驯化(5℃)滞育维持过程中的表达动态。与非滞育组相比,滞育起始阶段Gst基因相对表达量显著下调,AK和Tpi基因表达量无显著差异。在热驯化期间,仅Tpi基因表达量下调;冷驯化期间,3个基因表达量均上调,且呈逐渐上升的趋势。低温滞育期间mRNA的半衰期延长,mRNA转录逐渐积累,滞育过程和温度均影响亚洲玉米螟胁迫相关基因的表达。(3)了解越冬期间滞育幼虫的AK、Gst和Tpi基因表达的时间动态。分析越冬滞育幼虫AK、Gst和Tpi基因的表达量,亚洲玉米螟幼虫越冬期间AK、Gst和Tpi基因表达量均呈上调,且与越冬环境温度呈负相关性。表明能量代谢和抗氧化反应作用于亚洲玉米螟滞育幼虫的低温胁迫过程。
