木薯质体分裂相关基因ftsZ2家族的克隆及功能分析

【摘要】:本研究针对木薯淀粉品质改良问题,提出通过调控质体分裂元件,影响库器官中淀粉体分裂来改变淀粉粒形态、大小及充实程度的新思路。主要研究成果如下:完成ftsZ2基因家族全长片段在木薯的克隆,利用生物信息学预测功能及分析进化关系;利用FtsZ蛋白异常表达能严重影响大肠杆菌细胞分裂的特性,对内共生起源的木薯质体分裂基因MeftsZ2基因家族的功能进行初步鉴定;以我国木薯主栽品种SC8遗传转化体系,构建组成型和块根特异型启动子驱动的MeftsZ2-1基因的过量表达载体转化木薯,转化MeftsZ2-1基因的相关载体已得到经过分子检测的几十个转基因株系,并成功在田间种植,初步观察到苗期MeftsZ2-1转基因植株的叶柄和块根表皮颜色较野生型明显变红,表现出与质体发育相应表型的改变:此外还对多个品种淀粉粒度、淀粉体发育及淀粉粒发生过程进行了观察,为利用该途径改良木薯淀粉品质的新思路提供理论依据。
[Abstract]:In order to improve the quality of cassava starch, a new idea was put forward to change the shape, size and enrichment of starch grains by regulating the plastid division elements and affecting the amyloplast division in reservoir organs. The main research results are as follows: complete the cloning of ftsZ2 gene family in cassava, using bioinformatics to predict the function and analyze the evolutionary relationship; The abnormal expression of FtsZ protein can seriously affect the characteristics of cell division in E. coli, and the function of the MeftsZ2 gene family of cassava plastids derived from endosymbiosis was preliminarily identified. Based on the SC8 genetic transformation system of cassava main cultivar in China, the overexpression vector of MeftsZ2-1 gene driven by constitutive and tuber special type promoter was constructed to transform cassava into cassava. Dozens of transgenic lines have been obtained by molecular detection and successfully planted in the field. The color of petiole and root epidermis of MeftsZ2-1 transgenic plants at seedling stage is obviously red than that of wild type. The phenotypic changes corresponding to plastid development were also observed. In addition, starch granularity, amyloid development and starch granulation process of many varieties were observed, which provided theoretical basis for improving cassava starch quality by this way.
