调控CD163基因表达的miRNA鉴定及其在PRRSV感染中的作用分析

【摘要】:猪繁殖与呼吸综合征(PRRS)是由猪繁殖与呼吸综合征病毒(PRRSV)感染引起的一种危害大、控制难的猪传染性疾病。PRRSV必须通过特异性受体诱导才能入侵宿主细胞,而CD163就是其中一个关键受体。在获得靶向作用CD163基因miRNA的基础上,进一步研究发现过表达miR-181c、miR-4262可极显著抑制细胞内CD163 mRNA的表达(P0.01);且在Marc-145细胞中证实miR-181c、miR-23b、miR-4262均具有极显著抑制PRRSV增殖的功能(P0.01),其中miR-4262抑制效果最佳。3个miRNA抗PRRSV增殖的分子机制比较复杂,miR-181c、miR-4262可通过靶向抑制受体CD163发挥抗PRRSV增殖作用,而miR-23b是通过其他途径发挥作用。并首次发现miR-4262具有抑制PRRSV增殖的作用,但其具体的分子作用机制还有待进一步深入研究。
[Abstract]:Porcine reproductive and respiratory syndrome (PRRS) is a porcine infectious disease caused by porcine reproductive and respiratory syndrome virus (PRRSV) infection. PRRSv must be induced by specific receptors in order to invade host cells. CD163 is one of the key receptors. On the basis of targeting CD163 gene miRNA, it was found that over-expression of miR-181c,miR-4262 could significantly inhibit the expression of CD163 mRNA in cells (P0.01). Moreover, in Marc-145 cells, it was confirmed that miR-181c,miR-23b,miR-4262 could inhibit the proliferation of PRRSV significantly (P0.01), among which miR-4262 had the best inhibitory effect. The molecular mechanism of three miRNA against PRRSV proliferation was complex, and miR-181c, was found to be the most effective in inhibiting the proliferation of PRRSV (P0.01). MiR-4262 can inhibit the proliferation of PRRSV by targeting receptor CD163, while miR-23b plays a role through other pathways. It is found for the first time that miR-4262 can inhibit the proliferation of PRRSV, but its molecular mechanism needs to be further studied.
