马尔尼菲蓝状菌酵母相黑素生成和酪氨酸酶基因的关系

【摘要】:目的探讨马尔尼菲蓝状菌酵母相黑素生成和酪氨酸酶(TYR)基因的关系。方法将马尔尼菲蓝状菌标准株分别在含和不含L-DOPA的不同培养基中37℃避光培养3天,观察菌落形态及颜色变化,应用实时荧光定量PCR测定TYR基因mRNA的表达。结果(1)标准菌株接种在含和不含L-DOPA的SDA上菌落颜色无明显改变,未见明显黑素产生;(2)标准菌株接种在含有L-DOPA的BHI上菌落颜色深于接种在不含有L-DOPA的BHI的菌落颜色;(3)镜下观察标准菌株接种在含和不含L-DOPA的SDA,BHI上,其酵母样细胞的大小、形态基本一致,均未见被染成黑色。(4)TYR基因表达:接种在含L-DOPA的BHI的标准菌株,其TYR基因表达量高于不含L-DOPA的BHI组,差异有统计学意义(P0.05);含和不含L-DOPA的SDA中标准菌株TYR基因表达量均低于同组BHI组,差异有统计学意义(P0.05)。结论马尔尼菲蓝状菌在BHI中可以利用L-DOPA合成黑素;在BHI中添加外源性LDOPA时,马尔尼菲蓝状菌的TYR基因表达上调。
