染料木素对UVA所致成纤维细胞凋亡过程中Sirt1基因表达的影响

【摘要】:目的:探讨染料木素对UVA所致成纤维细胞凋亡过程中Sirt1基因表达的影响。方法:为了保证实验的准确性和可重复性,本实验将成纤维细胞分为7组,进行对照试验,其中A组为空白对照组(不添加染料木素,不照射UVA),剩余B至G组均照射UVA,具体划分是:B组为UVA照射对照组(不添加染料木素);实验组包括C组~G组,其实验药物的浓度分别为0.01μmol/L、0.1μmol/L、1μmol/L、10μmol/L和100μmol/L。采用MTT比色法检测成纤维细胞在UVA照射下染料木素对其增殖活性的影响,采用RT-PCR检测染料木素对Sirt1 m RNA的表达水平的变化。结果:与B(0.401±0.011)组比较,C~F(0.672±0.054、0.863±0.021、0.887±0.052、0.583±0.023)组细胞增值活性有明显升高,统计学上差异显著(P0.05),各实验组中D~E(0.863±0.021、0.887±0.052)组的细胞增值活性最高,统计学上差异显著(P0.05);与B(0.713±0.019)组比较,C~F(0.864±0.062、1.026±0.065、1.096±0.054、0.876±0.020)组Sirt1表达显著升高,统计学上有显著差异(P0.05),其中E组的Sirt1表达量显示最多,统计学上差异显著(P0.05),而B组与G(0.704±0.029)组相对照,Sirt1 m RNA的表达量无统计学差异(P0.05)。结论:染料木素在一定浓度范围内(0.01μmol/L~10μmol/L)对UVA致成纤维细胞的凋亡有防护作用,并且对成纤维细胞的Sirt1 m RNA的表达有显著增强作用,其中在浓度为0.1μmol/L~1μmol/L内保护作用最强。
