文心兰铁氧还蛋白基因克隆定位及表达分析
[Abstract]:A full-length 989bp cDNA sequence of ferredoxin gene, named OnFd (the length of open reading frame of accession number KX461907). OnFd gene was 465bp), was cloned from 'Cherry' Wenxin orchid by RACE-PCR, and it was predicted that it could encode 154amino acids. The results of gDNA and cDNA sequence alignment showed that OnFd gene did not contain introns. Bioinformatics analysis showed that OnFd had a typical [2Fe-2S] domain, and homology analysis showed that the similarity between OnFd and corn Fd3 was the highest (64.29%). The results of protein subcellular localization showed that OnFd was located in chloroplast. Real-time fluorescence quantitative PCR showed that the expression of OnFd gene was the highest in flowers, followed by leaves and roots, and the lowest in pseudo-bulb. The study of inoculated pathogenic bacteria showed that the expression of OnFd gene in all parts of Euphorbia pulcherrima increased significantly after inoculated with soft rot, especially after 1 day of inoculating pathogen. The expression of 2.83 脳 3.98 times as much as that of healthy plants in five susceptible stages. Studies have shown that OnFd gene may play an important role in the response of Wenxinlan to soft rot resistance.
