小鼠脾脏基质和B淋巴细胞的增龄性变化及同种异基因骨髓间充质干细胞移植对其的影响

【摘要】:研究目的1.检测BALB/c小鼠脾脏基质的增龄性变化,探讨β-catenin信号分子在脾脏基质老化中的作用。2.检测BALB/c小鼠脾脏B淋巴细胞比例、B淋巴细胞亚群比例和增殖、凋亡、周期相关基因mRNA表达水平的增龄性变化,探讨基质老化对B细胞老化的影响。3.探索C57BL/6J小鼠来源的同种异基因骨髓间充质干细胞移植对BALB/c、鼠脾脏基质和B细胞老化的影响。方法1.将雌性BALB/c小鼠分别按照2-4月龄、8-10月龄和18-20月龄定义为青年组、中年组和老年组。苏木精-伊红染色观察不同年龄组小鼠脾脏的组织学特征;天狼星红染色检测脾脏中胶原分布情况;免疫组织化学法检测脾脏collagen I、 elasti、fibronectin的蛋白表达情况。2.实时荧光定量PCR (Real-time PCR)法检测不同年龄组BALB/c小鼠脾脏基质成分agrin、college Iα1、collegen Ⅲα1 collegen Ⅳα1、collegen Ⅵα1、elastin、 fibrillin1、fibronectin 1、fibulin1、lamininα5、MMP-2的mRNA表达水平,p-catenin信号分子及其上游调控子DVL2、 DVL3、CK1、GSK-3β,下游转录子TCF4,靶基因PPARδ、 Axin2、c-myc、cyclinD1的mRNA相对表达量,蛋白质印迹法(Western blot)检测P-catenin和CK1的蛋白表达情况,Real-time PCR法检测B细胞标记CD19的mRNA相对表达量。3.制备脾脏单细胞悬液,流式细胞术检测不同年龄组BALB/c小鼠脾脏CD19+B淋巴细胞比例及B细胞中CD19+CD5+、CD19+CD24+、CD19+IgM+IgD-和CD19+IgD+细胞亚群的比例。4.流式细胞术分选、收集脾脏B淋巴细胞,Real-time PCR法检测B细胞中增殖相关基因Ki-67,凋亡相关基因Bax、Bcl-2、Caspase3、Caspase8,细胞周期相关基因cyclinD1、cyclinE1、p16INK4a,抗原决定簇分子CD5、CD24, β-catenin信号分子及其上游调控子DVL2、DVL3、CK1、GSK-3β,下游转录子TCF4,靶基因Axinδ、Axin2、c-myc的mRNA相对表达量。5.骨片消化法分离、培养C57BL/6J小鼠骨髓间充质干细胞(BMSCs)。6.通过尾静脉向实验组小鼠体内注射BMSCs,每2周注射一次,连续注射4次,单次注射量1x106个细胞(悬于100 μl PBS),末次注射后2周取材,重复检测上述脾脏基质和B细胞老化相关指标的变化。结果第一部分1.老年组小鼠脾脏组织结构紊乱,含铁血黄素颗粒和I型胶原沉积较青年组增加。2.老年组小鼠脾脏基质成分agrin、collagen Iα1、collagen Ⅲα1、collagen Ⅳα1、elastin和fibrillin 1的mRNA相对表达量显著低于青年组小鼠(P0.01或P0.05);collagen Ⅵα1、fibronectin 1、fibulin 1、laminin α5和MMP2的mRNA相对表达量与青年组小鼠无统计学差异。3.老年组小鼠脾脏β-catenin信号分子及其上游调控子CK1的mRNA和蛋白表达水平均低于青年组(P0.05或P0.01),下游靶基因PPARδ、cyclin D1、CD44的mRNA表达水平显著低于青年组(P0.01或P0.05);其他调控子DVL2、DVL3、GSK-3β,下游转录子TCF4,靶基因Axin2的mRNA相对表达量均值低于青年组,但未达到统计学差异(P0.05)。4.经过4次C57BL/6J小鼠来源的同种异基因BMSCs移植后,老年组BALB/c小鼠脾脏基质成分collagen Ⅲα1的mRNA表达显著上调(P0.01), collagen Ⅳα1和fibronectin 1的mRNA表达下调(P0.05和P0.01); β-catenin信号分子的下游转录子TCF4和靶基因CD44 mRNA表达上调(P0.05),其余相关基因表达无明显变化。第二部分1.不同年龄组BALB/c小鼠脾脏中B淋巴细胞比例无显著差异,老年组小鼠脾脏B细胞中CD19+CD5+、CD19+IgD+细胞亚群比例显著高于青年组(P0.01),CD19+CD24+、CD19+IgM+IgD'细胞亚群比例显著低于青年组(P0.01)。老年组脾脏B细胞中CD24、Ki-67的mRNA表达水平显著低于青年组(P0.01),CD5、Bcl-2、caspase 8和p16INK4a的mRNA表达水平显著高于青年组(P0.05或P0.01), Bax、caspase 3、cyclinD1和cyclinE1的mRNA表达水平与青年组无统计学差异。2.不同年龄组BALB/c小鼠脾脏B细胞中β-catenin信号分子及其上游调控子DVL2、DVL3、CK1,下游靶基因PPARδ、CD44、Axin2的mRNA表达水平无统计学差异。3.经过4次C57BL/6J小鼠来源的同种异基因BMSCs移植后,老年组小鼠脾脏B细胞比例和CD19的mRNA表达水平显著升高(P0.01);B细胞中CD19+CD5+、 CD19+CD24+和CD19+IgD+亚群比例显著降低(P0.01或P0.05);Ki-67和p16INK4a的mRNA表达水平上调(P0.01), CD5、CD24和Caspase 8的mRNA表达水平下调(P0.05),其余基因表达无明显变化。结论1.小鼠脾脏基质老化表现为多种细胞外基质成分的mRNA表达水平下调。2. β-catenin信号分子对脾脏基质老化有重要作用。3.随着年龄增加,小鼠脾脏B淋巴细胞比例维持稳定,但增殖活性降低,部分亚群比例发生改变,可能与基质老化有关。4.同种异基因骨髓间充质干细胞移植可以影响部分老化相关的脾脏基质和B细胞改变。

The changes of B-lymphocyte proportion, B-lymphocyte subpopulation ratio and proliferation, apoptosis and cycle-related gene mRNA expression level in BALB/ c mice were detected, and the effect of matrix aging on the aging of B cells was discussed.
